Endogenous production of superoxide by rabbit lungs: effects of hypoxia or metabolic inhibitors

Abstract: We find spontaneous light emission from isolated Krebs-Henseleit-perfused rabbit lungs when the light-emitting super-oxide trap lucigenin is added to the perfusate. Lucigenin light emission appears to be specific for superoxide anion, because light emission from the lung caused by a superoxide-generating system is abolished by superoxide dismutase but not by catalase or dimethylthiourea. We also studied the relative sensitivity of lucigenin photoemission to superoxide and to H2O2 in vitro. Lucigenin photoemiss… Show more

“…When compared to SOD, which can scavenge 02 only outside the cell, three of the test compounds (chrysin, DCI and NAS) were clearly more effective in inhibiting the PMA-induced formation of 02, suggesting that these antioxidants are also capable of scavenging intracellular O~-. This notion is supported by the finding that lucigenin is taken up by VSMC [24] and emits light upon reaction with intracellular 02 [25]. Another argument is that the antioxidants prevent the PMA-induced formation of O_~, because they interfere with the activation of PKC.…”

“…In contrast to DMTU, SOD is a large molecule (molecular weight: 15,100 as estimated by gel filtration [ 25 ] ) which does not readily gain access to the intracellular compartment (26,27) and remains in the extravascular space and lymphatics (28 …”

“…While ROS appears to exert some regulatory function on HIF-1, there is still debate as to whether ROS levels are increased [62,66,67] or decreased [68][69][70] during hypoxia. Contradictory results may occur due to differences in cell type, mode of generating hypoxia, oxygen levels and assays used to measure ROS.…”
