Enhanced cardiac angiotensinogen gene expression and angiotensin converting enzyme activity in tachypacing-induced heart failure in rats

Abstract: The aim of the study was to analyze changes in myocardial angiotensinogen gene expression and myocardial angiotensin converting enzyme activity in slowly progressing low-output failure. In adult, male Wistar rats, acute ventricular tachypacing by 610 to 620 impulses per minute lowered end-diastolic external diameter of the left ventricle by 2.6% (p less than 0.01), but did not lower cardiac output or abolish coronary reserve, since left-ventricular subendocardial blood flow of paced rats increased under dipyri… Show more

“…Activation of the cardiac RAS has been demonstrated to occur after myocardial infarction (13,14). This activation involves an increased transcription of the gene encoding the ACE mRNA (14)(15)(16).…”

“…Activation of the cardiac RAS has been demonstrated to occur after myocardial infarction (13,14). This activation involves an increased transcription of the gene encoding the ACE mRNA (14)(15)(16). The assessment of cardiac RAS, however, has been usually performed in chronically infarcted individuals with variable degrees of heart failure (13,17).…”

“…Recent studies have shown that the cardiac expression of angiotensinogen and ACE is increased in experimental heart failure ( 12,13,17,18). Cardiac ACE activity as well as the rate of angiotensin-II production is enhanced in hypertrophied rat hearts (12) and associated with altered diastolic properties possibly contributing to the vulnerability of diastolic function in hypertrophy during hypoperfusion ( 19).…”

“…Some authors have reported a transient increase of AOGEN mRNA in myocardial infarction and failure (175). A doubling of AOGEN gene expression as compared to control rats has been described in ventricular myocardium during cardiac chronic tachipacing (176). In chronic heart failure induced by ligation of the left coronary artery in rats an enhancement of ACE mRNA was found, which correlated with the cardiac ACE activity, but not with its activity in other organs (177).…”
