Enhanced Survival of Neural Cells Embedded in Hydrogels Composed of Collagen and Laminin-Derived Cell Adhesive Peptide

Abstract: To develop biomaterials that serve to improve the survival of neural cells transplanted into central nervous tissues, type I collagen-based hydrogels were prepared as a cell carrier. The hydrogels were modified with a laminin-derived peptide that is known to have an affinity for alpha3beta1 integrin, to transduce antiapoptotic signaling in embedded cells. For the modification of collagen, the peptide was fused to the N- or C-terminus, or both termini of a collagen-binding polypeptide domain by means of recombi… Show more

“…Several non-RGD peptides have been reported to promote NSC proliferation or differentiation, such as the laminin-derived PPFLMLLKGSTR [48] and IKVAV [7, 49] peptides. However, in previous work from our group, the IKVAV peptide failed to induce adhesion or proliferation of rat hippocampal NSCs when grafted to a polymer surface [25].…”

“…[18,22] Table 1). In cell encapsulation systems, although there are currently growing attempts in mimicking the effects of physiological ECM by introducing various synthetic and natural ECM materials into synthetic polymers, [5,[23][24][25] no reported studies to date have addressed the impact of ECM components properties on synthetic hydrogel characteristics, especially in terms of permeability. The current research investigates biosynthetic hydrogels fabricated via incorporation of heparin and gelatin into a PVA network.…”

“…The cell‐binding domains were chosen to target major integrins expressed by skin‐derived primary human epidermal keratinocytes. For example, the alpha3beta1integrin expressed by keratinocytes, has been shown to bind the PPFLMLLKGSTR sequence the laminin‐5 alpha3 chain globular domain 3 (LG3) . Both the PHSRN and RGD peptides as well as full length fragments of the 9 th and 10 th domains of fibronectin (FN910) have also shown to increase cell‐matrix interaction of keratinocytes via the alpha5beta1 integrin .…”
