Enhanced VWF biosynthesis and elevated plasma VWF due to a natural variant in the murine Vwf gene

Abstract: Both genetic and environmental influences contribute to the wide variation in plasma von Willebrand factor (VWF) levels observed in humans. Inbred mouse strains also have highly variable plasma VWF levels, providing a convenient model in which to study genetic modifiers of VWF. Previously, we identified a major modifier of VWF levels in the mouse (Mvwf1) as a regulatory mutation in murine Galgt2. We now report the identification of an additional murine VWF modifier

“…40 Whereas VWF levels in healthy humans range ;3-fold (from ;5 to ;15 mg/mL), 41 VWF levels among inbred mouse strains vary even more widely (.10-fold). 30 This latter variation is due to differences in VWF synthesis/secretion and clearance among mouse strains. 28,30 Genetic differences between the C57BL/6J and 129S1/SvImJ strains likely mice (C57BL/6J background) compared with similarly transfused HA mice (129S1/SvImJ background).…”

Because the same source of transfused VWF was used in both experiments, these data suggest that the ;2.5-fold difference in steady-state VWF levels between these strains 42 is primarily from an underlying clearance mechanism, rather than a difference intrinsic to the VWF protein itself.…”

“…22 Considerable variability in VWF levels has been noted in various strains of mice. 25 In contrast, strain-dependent variation of ADAMTS13 has not been described. In our effort to develop animal models for investigation of ADAMTS13 function in vivo, we discovered that mouse strains differed markedly in their plasma ADAMTS13 activity levels.…”

“…25,34 Nevertheless, variability in VWF levels does not correlate with the risk of thrombosis in mice with ADAMTS13 deficiency. 35 The present study reveals that mouse strains also differ considerably in their ADAMTS13 activity levels.…”

“…This represents a greater level of divergence between B6 and WSB, 2 strains that are thought to be primarily mus musculus domesticus in origin, versus A/J and CASA/RkJ (the strains used to identify Mvwf2-4), which are mus musculus domesticus and mus musculus castaneus, respectively. 17 To evaluate potential cis-regulatory differences in mRNA expression or stability, lung mRNA was isolated from (B6 ϫ WSB) F1 mice and subjected to RT-PCR for Vwf mRNA, followed by primer extension analysis that distinguishes the B6 and WSB alleles because of an exonic SNP ( Figure 5A). A similar approach has been successful in distinguishing expression from maternal and paternal alleles in cases of human VWD.…”
