Enhancement of Endometrial Receptivity by Cnidium officinale through Expressing LIF and Integrins

Abstract: Improvement of endometrial receptivity is necessary for successful embryo implantation, and its impairment is associated with female infertility. In this study, we investigated the effect of the roots of Cnidium officinale Makino (CoM) on endometrial receptivity in both in vitro and in vivo model of embryo implantation. We found that CoM enhanced the adhesion of JAr cells to Ishikawa cells by stimulating expression of leukemia inhibitory factor (LIF) and integrins. In addition, blocking of LIFR using hLA or ne… Show more

“…Thus, LIF and integrin ανβ3 are used as indicators of the establishment of uterine receptivity. In our study, the downregulation of GLUT4 resulted in the defective glucose uptake of endometrial epithelial cells, which further led to impaired endometrial receptivity by inhibiting pinopode formation and the expressions of endometrial receptivity markers, LIF and integrin ανβ3, and then impaired implantation (Franasiak et al, 2014;Chung et al, 2019). In embryo transfer experiments, pretreatment of one side of the mouse uterine horns with GLUT4-siRNA also resulted in a reduced implantation rate, even if excellent embryos were selected for transfer.…”

“…Although assisted reproduction techniques (ART) have been widely used for the clinical treatment of infertility resulting from benign gynecological disorders or primary unexplained infertility, subsequent pregnancy rates remain unsatisfactory [4,7]. In traditional Eastern medicine, herbal medicines, acupuncture, and moxibustion are used to treat female infertility [8,9], and recently, due to accumulated scientific evidence, herbal medicine has emerged as an effective, complementary, alternative medicine for improving endometrial receptivity [10,11].…”

“…Although this prescription is used clinically to treat female infertility, experimental evidence obtained from in vitro and in vivo laboratory studies is lacking [ 6 ]. Mifepristone (also known as RU486) is a potent antiprogesterone agent, which can be used orally to induce abortion [ 16 ] or as an effective emergency contraceptive [ 17 ], and has often been used to induce embryo implantation failure or polycystic ovary syndrome in experimental animal models [ 10 , 18 , 19 ]. In the present study, we investigated the effects of a BCB water extract on endometrial receptivity and embryo implantation under normal conditions and in an RU486-induced mouse model of implantation failure.…”

“…Furthermore, it decreased production of human chorionic gonadotropin (hCG), which in turn results in decreased production of progesterone by the corpus luteum. RU486 has been often used to induce embryo implantation failure or polycystic ovary syndrome in experimental animal models [11,[17][18][19]].…”
