Enhancement of the antifungal activity ofBacillus subtilisF29-3 by the chitinase encoded byBacillus circulans chiAgene

Abstract: Bacillus subtilis F29-3 is an antagonistic bacterium against a wide range of fungal species. In order to determine the effect of chitinase on the antifungal activity of B. subtilis F29-3, a 2.4-kb DNA fragment containing the chiA gene of Bacillus circulans WL-12 was ligated into a shuttle vector pHY300PLK and transformed into B. subtilis F29-3. A bioassay conducted on the culture supernatant showed that, in comparison to the B. subtilis control strain, B. subtilis F29-3 expressing the chiA gene exhibited a gre… Show more

“…Shanmugaiah et al (2008) reported that among the 39 chitinolytic bacteria which were isolated from rice rhizosphere, only 11 isolates produced zone of clearance over 0.5 cm on chitin colloidal agar. Chitinase production was reported in different species of Bacillus such as B. amyloliquefaciens (Wang et al, 2002), B. cereus (Huang et al, 2005), B. circulans (Chen et al, 2004), B. licheniformis (Waldeck et al, 2006), B. megaterium (Donderski and Brzezinska, 2005), B. stearothermophilus (Sakai et al, 1994), B. subtilis (Wang et al, 2006), B. thuringiensis sub sp. aizawai (De la Vega et al, 2006), B. thuringiensis sub sp.…”

“…739, and in studies with B. amyloliquefaciens MET0908, with the aid of scanning electronic microscopy, the lytic activity of a β -1,3-glucanases on the Colletotrichum lagenarium hyphae was observed. Also, genetic engineering studies focused on the increase of antifungal activity of Bacillus strains have successfully achieved the insertion of codifying genes into lytic enzymes, such as gene ChiA of B. subtilis F29-3 in B. circulans, obtaining a more aggressive phenotype against Botrytis elliptica (Chen et al, 2004). In addition, Zhang et al (2012) significantly increased the antifungal activity of Burkholderia vietnamiensis P418 in the face of different fungal phytopathogens by the chromosomal insertion of the gene Chi113, from a B. Subtilis strain.…”

“…Further, mutants (W81M3 and W81M4) which have protease overproduction capacity exhibited noticeably improved biocontrol potential (Dunne et al 2000 ) . Similarly cloning of chitinase gene chiA of B. circulans WL-12 in strain B. subtilis F29-3 has shown that in comparison to the B. subtilis control strain, chiA expressing B. subtilis F29-3 exhibited a greater inhibition of spore germination of Botrytis elliptica (Chen et al 2004 ) . Hence it may be concluded that pathogen colonization and parasitism is a key mechanism adopted by many rhizobacteria to inhibit the fungal pathogen and such organisms have an edge over others in terms of effective control of the target pathogen (Fig.…”
