Enrichment and Detection of Tyrosine‐Nitrated Proteins

Abstract: Nitrotyrosine is a post-translationally modified amino acid with distinctly different properties than tyrosine or any other of the genetically encoded amino acids. Detecting proteins containing nitrotyrosine is the first step towards a better understanding of the role of nitrotyrosine in health and disease. Moreover, quantifying the extent of nitrotyrosine and determining its location in a protein forms the basis for a better understanding of the effect of tyrosine nitration on biological function. “…MS-based analyses of tyrosine-nitrated peptides include several different separation and enrichment procedures (see for instance Lee et al, 2009;Draguşanu et al, 2010;Draguşanu, Petre, & Przybylski, 2011;Dremina et al, 2011;Prokai-Tatrai, Guo, & Prokai, 2011). Some of these procedures have been reviewed recently (Nuriel et al, 2008;Abello et al, 2009;Dekker et al, 2012). For a brief summary see Table 6.…”

“…Thus, the combination of proteomic techniques (two-dimensional polyacryamide gel electrophoresis) followed by immunoblotting or immunoprecipitation, gel tryptic digestion and mass spectrometry have become powerful tools to develop nitroproteome studies in higher plants. Although there might be some technical problems like those due to the unspecific recognition of the antibody used against nitrotyrosine, the low number of nitrotyrosines in a given protein, the low abundance of the nitrotyrosine-containing proteins, and that the isolated protein occasionally does not match in the protein database, it must be pointed out that these approaches have allowed establishing a basic background of knowledge in this research area (Dekker et al, 2012). In any case, after a specific protein has been identified as a putative nitration target, a necessary additional step is to identify the nitration site(s) within the protein quaternary structure by MALDI-TOF MS and LC-MS/MS (Ytterberg and Jensen, 2010).…”

“…Currently, an enrichment technique followed by MS is the most powerful approach for detecting nitropeptides. Enrichment procedures include use of either 3-NT specific antibodies or chemical derivation that reduces a nitro group to an amine group (17)(18)(19). Most previous studies aimed at methodology development and used in vitro overnitrated samples to profile artificially nitrated instead of endogenously nitrated proteins.…”
