Enrichment of Human Stem-Like Prostate Cells with s-SHIP Promoter Activity Uncovers a Role in Stemness for the Long Noncoding RNA H19

Abstract: Understanding normal and cancer stem cells should provide insights into the origin of prostate cancer and their mechanisms of resistance to current treatment strategies. In this study, we isolated and characterized stem-like cells present in the immortalized human prostate cell line, RWPE-1. We used a reporter system with green fluorescent protein (GFP) driven by the promoter of s-SHIP (for stem-SH2-domain-containing 5¢-inositol phosphatase) whose stem cell-specific expression has been previously shown. We obs… Show more

“…Furthermore, a recent study of prostate and breast human cell lines revealed a putative role of stemness for H19 , since its expression was positively correlated with stem cells markers and pluripotency factors. H19 was also found to control the expression of two major pluripotency factors, Oct4 and Sox2, which apparently regulate H19 in an amplification feedback loop [ 87 ].…”

“…H19 expression has been shown to positively correlate with the level of stem cell genes and pluripotency factors in various tumor types (Bauderlique‐Le Roy et al , 2015; Li et al , 2016; Peng et al , 2017; Zeira et al , 2015). Comparison of the PIM‐i‐resistant (SUP‐T1 and CUTLL1) and PIM‐i‐sensitive (HSB‐2 and DU.528) T‐ALL cell lines revealed significantly elevated SOX2 expression in sensitive vs resistant T‐ALL cells (Fig.…”

“…We have previously shown that s-SHIP-GFP promoter reporter tracks subset of human prostate RWPE-1 cells enriched in stem cell characteristics [ 45 ]. s-SHIP is also expressed in numerous cancer cell lines and xenograft–maintained human prostate cancers ( Supplementary Figure 3A ) which represent relevant models of prostate cancer [ 46 – 48 ].…”
