Enumeration of Nucleated Red Blood Cells with the ADVIA 2120 Hematology System: An International Multicenter Clinical Trial

Abstract: The accurate, reproducible, and timely reporting of nucleated red blood cells (NRBC) is an important function of the clinical hematology laboratory. We used 960 samples from 5 worldwide sites to evaluate a new NRBC enumeration method for the ADVIA 2120 Hematology System. The method showed excellent correlation with microscopy (r = 0.93). Sensitivity and specificity for the presence of NRBC for all samples analyzed was 77.3% and 74.6%, respectively. Almost all false negative samples were at NRBC counts Show more

“…[13,14] Because infectious conditions are associated with increased immature granulocyte levels, several investigators have examined whether this level can be used to predict the development of sepsis. [15,16] On the other hand, the serum myeloperoxidase index (MPXI) is a new index of the level of serum myeloperoxidase (MPO), which is a newly observed inflammatory marker.…”

“…DNI is calculated by an automatic counter (ADVIA 120; Siemens Healthcare Diagnostics) using myeloperoxidase (MPO) channels and nuclear lobularity channels in the test system [12]. The difference between the leukocyte differentials assayed in the two channels is designated as the DNI, which correlates with the fraction of immature granulocytes in peripheral blood calculated by manual counting.…”

“…[19][20][21] DNI is assessed by an automated blood cell analyzer and tests required for the DNI can be performed at the same time as routine complete blood count (CBC) test. 22 It measures the fraction of immature granulocytes in the circulation.…”
