Enzyme electrophoresis in characterizing the causative organism of Gambian trypanosomiasis

Abstract: Trypanosoma (Trypanozoon) brucei includes three morphologically identical subspecies which are poorly defined by clinical behaviour; T. b. brucei does not infect man, whereas T. b. rhodesiense causes an acute, and T. b gambiense a chronic, disease. Thirty-three isolates of the complex, each of which had previously been identified on clinical or other criteria, were compared by the electrophoretic patterns of two trypanosomal enzymes, alanine aminotransferase (ALAT) and aspartate aminotransferase (ASAT). One pa… Show more

“…are among some of the most wellstudied agents of parasitic diseases known today. In consequence, some of these organisms have long been the subject of investigation and speculation with regard to their evolutionary history (Léger, 1904;Wallace, 1966;Hoare, 1972;Vickerman, 1994) and, indeed, trypanosomatids were among some of the first parasites to be characterised with the new methods of isoenzyme typing developed in the 1970s (Godfrey & Kilgour, 1976;Miles et al, 1977). For many kinetoplastid groups (notably those of medical importance), the findings of early studies have since been superseded by results from more recent studies, which, in general, due to the techniques used, e.g.…”

“…Parasites were harvested by centrifugation and pellets stored at -70ºC. Before electrophoresis, pellets were mixed with an equal volume of enzyme stabilizer (Godfrey & Kilgour 1976) and left on ice for 20 min. They were then centrifuged at 11,000 g at 4ºC for 10 min.…”
