“…Miniaturized versions have been used as scanning probes to monitor the glucose and lactate metabolism of single cells 42,43 . Flow-through-type sensor systems have been devised to analyze cell culture conditions and metabolites in liquid samples downstream of cell cultures [44][45][46][47][48][49][50][51] . Silicon-based sensor chips for multi-parameter online monitoring have been mounted in a perfused cell culture unit 52 , and multiple sensing electrodes have been directly incorporated into transparent microfluidic systems to measure the glucose consumption of single cardiac cells 53 or the lactate production of cells of various types 54 and, combined with optical read-out, for monitoring cancer cell metabolism 55 .…”

“…These LOC devices are good examples of how changes at single cell level can be monitored upon different stimuli using electrochemical sensing. Several of other electrochemical LOC devices implemented for the detection of cell-secreted lactate were also reported [96,98,99,112,113]. Many of these electrochemical lactate biosensors are amperometric and operate at high applied potentials that vary between 0.45 V to 0.70 V and used to record the current changes caused by the electrochemical oxidation of H 2 O 2 and thus these biosensors suffer from low selectivity.…”

“…64,7174 Frey et al fabricated a disposable PDMS cartridge for the measurement of glucose and lactate in solution featuring on-line adjustment of the linear response range and sensitivity of the biosensor 68. The same group later developed a novel cartridge sensor for cell-culture applications featuring a limit of detection of less than 5 nM 69…”

“…The flexibility provided by the enzyme functionalization has led to the realization of a large variety of biosensors for the analysis of cell cultures and metabolites in liquid samples . Typical examples for cell culture applications include glucose and lactate electrochemical biosensors, the sensing electrodes of which are functionalized with glucose oxidase or lactate oxidase. Upon interaction of the oxidase enzyme with the target substrate, hydrogen peroxide is produced, which can then be detected using an amperometric detection scheme. Using an SECM featuring glucose or lactate enzymatic biosensors at the tip, Ciobanu et al were able to measure the glucose consumption of individual fibroblasts and the lactate secretion of single cancer cells cultured in standard Petri dishes .…”
