Enzymetically Regulating the Self‐Healing of Protein Hydrogels with High Healing Efficiency

Abstract: Enzyme-mediated self-healing of dynamic covalent bond-driven protein hydrogels was realized by the synergy of two enzymes, glucose oxidase (GOX) and catalase (CAT). The reversible covalent attachment of glutaraldehyde to lysine residues of GOX, CAT, and bovine serum albumin (BSA) led to the formation and functionalization of the self-healing protein hydrogel system. The enzyme-mediated protein hydrogels exhibit excellent self-healing properties with 100% recovery. The self-healing process was reversible and ef… Show more

“…Injectable chemically cross‐linked hydrogels are generated by introducing covalent linkages between polymer chains through Schiff's base reactions, photo‐irradiation, enzyme, Michael‐type addition reaction, disulfide formation, and Diels–Alder reaction . Once intermolecular cross‐linking has occurred with adequate cross‐linking density due to the generation of new covalent bonds, three‐dimensional (3D) network structures may be formed.…”

“…A direct and simple way of incorporating proteins into cross‐linked structures is the use of protein molecules as cross‐linkers. Amino and carboxylic acid groups on proteins were used directly in the cross‐linking reactions, or modified with the active functional groups for further cross‐linking reactions . For example, Yamauchi and co‐workers synthesized hydrogels based on a reaction between the NHS‐modified alginic acid (AL) and the HSA macro‐cross‐linker .…”

“…Furthermore, some unique properties of self-healing materials are quite intriguing to in vivo applications. One instance is a bovine serum albumin (BSA) hydrogel regulated by glucose oxidase (GO X ) and catalase (CAT), in which recovery is stimulated by the presence of glucose and 100% healing efficiency could be obtained within 5 h at room temperature [25]. Due to the broad applications of selfhealing materials, this section mainly focuses on summarizing and discussing various applications of self-healing materials in the biomedical field.…”
