Enzymic synthesis of fatty esters by hydrophobic lipase derivatives immobilized on organic polymer beads

Abstract: Lipase fromCandida rugosa was modified with several hydrophobic modifiers before being adsorbed onto organic polymer beads. The effects of different enzyme modifiers, supports, solvents, reaction temperatures, fatty acids, and alcohols on the activity of the immobilized enzyme were investigated. The immobilized lipases were good biocatalysts for esterification reactions in organic solvents. They exhibited high activities in all solvents tested, including polar solvents. The activity seemed to depend on the typ… Show more

“…Lipases are also able to undergo such "interfacial activation" in the presence of other hydrophobic interfaces: lipid micelles (the natural substrate) (Basri et al, 1995;, hydrophobic supports (Al-Duri et al, 1995;Balcão and Malcata, 1998;Balcão et al, 1996;Balcão et al, 1998aBalcão et al, , 1998bBastida et al, 1998;Fernández-Lafuente et al, 1998;Reetz, 1997;Sabuquillo et al, 1998;Sugiura and Isobe, 1975;1976a;1976b;1977) and hydrophobic proteins (fungal hydrophobins) (Palomo et al, 2002).…”

“…Because biocatalysts have high specific activity and a low impact on the environment, they have become increasingly important for industries. For example, immobilized lipases have been used as catalysts for reactions involving the biomodification of triglycerides (BASRI et al, 1995).…”

“…A crescente ênfase no uso de biocatalisadores devido às suas propriedades favoráveis de reação tais como condições amenas e ambientalmente compatíveis e sua alta especificidade, tem resultado num aumento do uso de enzimas imobilizadas. Isto vem ocorrendo pois as interações entre o suporte e a enzima podem alterar favoravelmente as propriedades físicas e químicas da enzima e, geralmente, uma nova propriedade é gerada, que pode ser explorada para propostas industriais [4].…”
