The study was performed on 225 chicken (72 diseased and 153 freshly dead ones) from different 45 commercial broiler farms (1-30 days old) at Dakahlia Governorate for mycotic infection. Samples were taken from these chickens (lung; air sac; crop; liver and brain from each chicken) after clinical and postmortem examination for mycological examination. The results revealed that, fungi were isolated from 503 positive samples (44.7%); represented as 119 positive samples (10.6%) from diseased chickens and 384 positive samples (34.1 %) from freshly dead ones.651 fungal isolates (moulds and yeasts) were isolated from 1125 samples, where 219(33.6%) were isolated from lung samples followed by 199(30.6%); 119(18.3%); 91(14.0%) and 23(3.5%) from air sac, liver, crop and brain samples respectively. Moreover, A. fumigatus was the most isolated one 141(21.7%) followed by A. flavus 126(19.4%); A. niger 111(17.1%); Rhizopus spp. 67(10.3%); C. albicans 58(8.9%); Mucor spp. 42(6.5%); Penicillium spp. 32(4.9%); A. terreus 20(3.1%); Fusarium spp.19 (2.9%); C. krusei 13(2.0%); C. tropicalis 9(1.4%); A. candidus 9(1.4%) and finally A. ochraceus 4(0.6%). PCR using fungus-specific universal primer pairs (ITS1 and ITS4) was used for identification and genotypic characters of Aspergillus species (A. fumigatus; A. flavus and A. niger).They were able to successfully amplify the ITS1-5.8S rDNA region of all tested Aspergillus isolates, providing a single PCR product of about 600 bp for all tested isolates PCR using fungus-specific universal primer pairs (ITS and RPS) was used for identification and genotypic characters of C. providing a single PCR product of about 109 bp .Sequence of the internal transcribed spacer 1 reigon (ITS-1) of isolated A. flavus Gene Bank accession number for studied nucleotide sequence (Bankit 1867303) is KM 983253. The sequences obtained for ITS-1 region were more than 98% identical to the corresponding GenBank sequences (accession no. DQ467968.1; KC994648.1; LN482516.1 and KP689246.1). Sequence of the internal transcribed spacer 1 region (ITS-1) of isolated C. albicans. Gene Bank accession number for studied nucleotide sequence (Bankit 1867955) is KM 983254. The sequences obtained for ITS-1 region obtained were more than 95% identical to the corresponding GenBank (accession no. M87288.1; gb|S71769.1|S71769; emb (AL033396.1) and AP006852.1).

Objectives: This study was designed to isolate, identify, and determine the prevalence of Aspergilli in commercial chicken in selected areas of Bangladesh. Materials and Methods: A total of 50 lung samples from suspected dead chickens, comprising broilers (n = 32) and layers (n = 18), aged between 5 days and 45 weeks, were collected from poultry farms located in the Gazipur district in Bangladesh. Fungi were primarily identified based on the colony morphology using potato dextrose agar (PDA). DNA was extracted from the suspected colonies. Aspegillus spp. was detected by genus-specific ASAP-1 and ASAP-2. Aspergillus spp. were then screened by polymerase chain reaction targeting Aspergillus flavus (FLA-1 and FLA-2), Aspergillus fumigatus (ASPU and Af3r), and Aspergillus niger (ASPU and Nilr). Results: The overall prevalence of Aspergillus spp. was 44% (n = 22/50; p < 0.05). Among the Aspergilli, A. flavus was detected in 10% (n = 5/50) of the samples. Similarly, A. fumigatus and A. niger were detected at 26% (n = 13/50) and 8% (n = 4/50) respectively. Three samples were associated with more than one fungus; two fungi (A. flavus and A. niger) were in two samples, and three fungi (A. flavus, A. fumigatus, and A. niger) were in one sample. Conclusion: Isolation and prevalence of Aspergillus spp. in commercial chicken were studied for the first time in Bangladesh.

Environmental Monitoring of Zoonotic Fungal Infection in Broiler Chickens: Novel Approach to Control using Nano-fungicide Composite

Control of fungal infections has not taken much attention compared to bacterial and viral pathogens inflicting significant economic losses to the poultry sectors as well as direct harms to human health due to their zoonotic implication. This study aimed to investigate the existence of fungal pathogens in broiler chicks dropping and their environment. As well, evaluate the efficiency of Terminator disinfectant (coco-benzyl-dimethyl ammonium chlorides and glutaraldehyde), nano copper oxide (CuO NPs), and Terminator/ nano copper oxide (Terminator/CuO NPs) on the fungal growth inhibition to control resistant fungus to antifungal agents. All samples (n= 320) were collected from chicks dropping, their environment (air, water, feeds, litter, drinkers, and feeders) as well, the attendant's hand swabs for isolation and identification of fungal pathogens. The susceptibility pattern of 65 strains of fungal isolates to antifungal agents, terminator disinfectant, and nanofungicide composites was determined by the disc diffusion assay and broth micro-dilution method. All isolates were highly resistant to voriconazole antifungal drugs, whilst Aspergillus fumigatus (A. fumigatus) was resistant (100%) to fluconazole. Furthermore, the sensitivity of Aspergillus terreus (A. terreus) and Penicillium corylophilum (P. corylophilum) was 0.0% to fluconazole, and amphotericin-B. Whilst the antifungal activity of Terminator/CuO NPs against fungal pathogens proved its lethal effect (100%) against all fungal isolates at 0.5 mg/mL compared to the efficiency of both Terminator at 1:50 and CuO NPs at 2.0 mg/mL was not exceeded 84.6% and 76.9%, respectively against all fungal strains. In conclusion, nano-fungicide is a promising product for the prevention of fungal pathogens in broiler chickens and their environment. The control of zoonotic resistant fungus using novel nano-fungicide composite (Terminator/CuO NPs) at 0.5 mg/mL concentration was efficiently achieved compared to nano copper oxide at 2.0 mg/mL.
