Epigallocatechin-3-gallate added after thawing to frozen dog semen: Effect on sperm parameters and ability to bind to oocytes’ zona pellucida

Abstract: Dog sperm cryopreservation is gaining importance both in breeding dogs for commercial purposes and for pet animals. Anyway, cryopreservation of mammalian spermatozoa, including dog ones, induces some negative effect on sperm fertility, leading to a lower use of this technique and limiting its widespread use. Therefore, studies to improve the quality of canine semen after cryopreservation could have a relevant impact on both the scientific advancement and the clinical practice. The aim of the present work was to… Show more

“…shown that the addition of EGCG can improve the motility and acrosome integrity of dog sperm, thereby improving the fertilisation ability of sperm (Bucci et al, 2019). Adding EGCG to improve sperm kinematics parameters may be due to the reduction of bacterial contamination caused by air exposure (Tsou et al, 2017).…”

“…Sperm motility is an important factor that affects subsequent insemination, and the integrity of the acrosome is also the key to whether the sperm can dissolve the radioactive crown and the transparent zone and successfully enter the ovum. Previous studies have shown that the addition of EGCG can improve the motility and acrosome integrity of dog sperm, thereby improving the fertilisation ability of sperm (Bucci et al, 2019). Adding EGCG to improve sperm kinematics parameters may be due to the reduction of bacterial contamination caused by air exposure (Tsou et al, 2017).…”

“…, addition of prostatic fluid after thawing may be appropriate to improve sperm motility [ 67 , 68 , 69 ]. In addition, sperm motility is reportedly improved by post-thawing addition of pentoxifylline [ 70 , 71 , 72 ], epigallocatechin-3-gallate [ 73 ], caffeine [ 71 , 72 ], and 2'-deoxyadenosine [ 71 , 72 ]. However, the fertility of treated spermatozoa after artificial insemination has not yet been assessed.…”

“…8,9 However, a recent study by Bucci et al demonstrated that EGCG supplementation to thawing medium failed to improve dog sperm quality or zona binding capacity. 10 As far as we know, there is still a lack of systematic research on the effects of EGCG on the IVM and IVF of porcine oocytes. Whether EGCG can be used as an effective antioxidant for porcine oocytes cultured in vitro remains to be explored.…”
