Epigallocatechin gallate induced apoptosis in Sarcoma180 cells in vivo: Mediated by p53 pathway and inhibition in U1B, U4-U6 UsnRNAs expression

Abstract: The aim of this study was to understand the mode of action of tea polyphenol epigallocatechin gallate (EGCG) in vivo. Swiss albino mice were treated i.p. with EGCG at two different doses i.e. 12-mg/kg body weight and 15-mg/kg body weight, for 7 days prior to inoculation of Sarcoma180 (S180) cells and continued for another 7 days. The growth of the S180, harvested 7 days after inoculation, was significantly reduced due to treatment with EGCG. The flowcytometric analysis of S180 cells, showed significant increas… Show more

“…Promotion of apoptotic cell death by EGCG in sarcoma 180 cells was induced through G2/M cell cycle arrest, down-regulation of Bcl-2 and c-myc, up-regulation of p53 and Bax and without changes in the expression of p21, p27, Bcl-x L , mdm2 and cyclin D1 [96]. Up-regulation of p21, p53 and Bax have also been reported in prostate cancer PC-3 cells together with increased levels of caspases-3, -9 and proteolytic cleavage of poly ADP-ribose polymerase (PPARc) [68].…”

“…The slides were washed with PBS and finally incubated with horseradish POD conjugated anti-fluorescein antibody and DAB as the chromogen for 30 mins at 37°C under humidified conditions. Stained slides were visualized and photographed under a bright field microscope (Manna et al, 2006).…”

“…The anti-tumor effect of flavonoids could also be exerted through the stimulation of apoptotic death, via activation of caspases (caspase-3, -9, and -8) and pro-apoptotic proteins (p53, p27 and Bax) as well as by inhibiting antiapoptotic proteins (Bcl-2 and Bid) and the release of cytochrome c [155][156][157][158].…”
