For the first time in the Russian Federation, using the example of the Vologda region of the North-West of the Russian Federation in pig farms by high-performance sequencing of amplicon libraries of fragments of the 18S rRNA gene obtained as a result of nested PCR, we have established parasitization of Cryptosporidium scrofarum in pigs of all age groups. The infection rate of animals kept in pig farms was 34%, in farms – 32.4%. Piglets that are fattening at the age of 13-24 weeks are most susceptible to infection. The analysis of the taxonomic affiliation of ASV carried out using phylogenetic analysis, supplemented by analysis using the blastn algorithm in the GenBank database, showed that in total, 10 ASV types (amplicon sequence variant) with high similarity to sequences deposited in GenBank as fragments of the Cryptosporidium scrofarum 18S rRNA gene are present in all the samples studied. It was found that the ASV1 and ASV2 types detected in various geographical regions of the world from Portugal and Great Britain to China, India and Australia were identified in all surveyed farms, although in significantly different quantities. The remaining ASVS belong to local populations of C. scrofarum subspecies. A unique sequence of the genus Cryptosporidium of type ASV 8 has been discovered, which can later be described as a new species. The nucleotide sequences we have discovered are unique. Each of them was deposited in GenBank with the assignment of identifiers (Sequence ID: OR649139, OR654022, OR654023, OR661243, OR661244, OR654051, OR654052, OR654083, OR654084, OR654106).

Cryptosporidiosis is a widespread parasitic disease of many species of domestic and wild animals, as well as humans, which is a significant problem in the field of veterinary medicine and medicine. Farm animals, in particular piglets, are most often susceptible to this disease, however, the species composition of representatives of the genus Cryptosporidium in this animal species in the Russian Federation has remained unknown to this day. For the first time in the Russian Federation, in the conditions of the North-West, on the example of the Vologda region in pig farms with industrial technology of maintenance, as well as in private farms using the latest molecular genetic techniques, namely, using high-performance sequencing of amplicon libraries of fragments of the 18S rRNA gene obtained as a result of nested (nested) PCR, we have established the parasitism of C. scrofarum in all age groups of the examined animals. The infection rate of animals kept in pig farms was 34% (51/150), in farms – 32.4% (81/250). The most susceptible to infection are animals that are fattening at the age of 13-24 weeks.

The purpose of the research is to study the contamination of environmental objects with Cryptosporidium spp. oocysts in industrial pig farm; desinfection against exogenous stages of Cryptosporidium spp. using Kenocox.Materials and methods. The studies were carried out on the basis of an industrial pig breeding complex in the Cherepovets district of the Vologda region in a pigsty for 120 heads, divided into 2 sectors, as well as in a pigsty for weaning pigs and a fattening pigsty for 1500 and 2000 heads respectively. For this, scrapings were taken from the floors of the pens, walkways, and walls of feeders and examined them for the presence of Cryptosporidium spp. oocysts. Then, in the nursery after transferring the piglets to the rearing section, desinfection was carried out. One sector was treated with Kenocox at a dose of 0.5 L per 1 m2 with an exposure of 2 hours, the other sector – with a hot 4% sodium hydroxide solution at a rate of 1 L per 1 m2 with an exposure of 3 hours. One day after the treatment, the oocyst contamination of Cryptosporidium spp. in environmental objects in both sectors was re-examined.Results and discussion. The highest contamination of environmental objects with Cryptosporidium spp. oocysts (floors and walls of pens, feeders, floors of aisles) was found in the brood pigsty, where all the examined objects were contaminated to varying degrees (19.4%). With an increase in the age of piglets and their transfer to pigsties for weaners, and later on for rearing, there is a decrease in the contamination of the premises in which these age groups of animals are kept (10.0 and 2.5% respectively). With the desinfection of the queen-shed, as the most contaminated by oocysts of Cryptosporidium spp. from the pig-breeding premises we examined, a significant effect was achieved with the use of Kenocox. Sodium hydroxide solution was found to be ineffective against Cryptosporidium spp. oocysts.
