Erectogenic and Aphrodisiac Effects ofButea frondosaKoenig ex Roxb. in Rats: Involvement of Enzyme Inhibition

Abstract: Butea frondosa Koenig ex Roxb. (BF) is traditionally used to manage male sexual disorders including erectile dysfunction (ED). Methanol extract of BF (bark) inhibited Rho-kinase 2 (ROCK-II) enzyme activity in vitro with an IC50 of 20.29 ± 1.83 μg/mL. The relaxant effect of methanol extract of BF (MEBF) was studied on phenylephrine precontracted corpus cavernosum smooth muscle (CCSM) isolated from young rats. The effect of MEBF treatment on sexual behaviour of both young (5 month) and aged (24 month) rats was a… Show more

“…Paraffin sections of tissue (5 mm thickness) were taken and Masson's trichrome staining was performed for determining smooth muscle:collagen level using a standard protocol. 13,19,20 Briefly, the tissues were hydrated and dipped in bouin's fluid for 12 h at 27 C. The tissues were then washed and rinsed in distilled water followed by staining in Wigards iron haematoxylene for 10 min. The tissues were subsequently washed and rinsed in distilled water followed by staining in Biebrich scarletacid fuchsin for 5 min.…”

“…Coordinated relaxation of penile smooth and penile arteries results in persistent penile erection required for sexual activity. 6,12,13,15,18 Interestingly, testosterone, a male sex hormone stimulates and maintains the NOS. [29][30][31] We estimated nitrite, the stable metabolite of NO in the serum of rats.…”

“…These were assessed for the number and gross morphology. 15 For sperm motility, an aliquot of 10 µL was placed in a hemocytometer chamber and analyzed under a light microscope. One hundred sperm were evaluated per animal and classified into motile and immotile.…”

“…The aphrodisiac property of many Indian medicinal plants which has been associated with an increase in the level of male hormone testosterone is known to control sexual function (Andersson, ; Andersson, ; Zvara et al , ; Putnam et al , ; Shabsigh, ; Sharma et al , ; Sharma et al , ). Peripherally, penile erection is regulated by contraction mediated by transmitters/modulators (noradrenaline, endothelin, angiotensin, Rho kinase, and arginase) and relaxation mediated by transmitters/modulators such as acetylcholine, NO, and adenosine (Andersson, ; Shabsigh, ; Andersson, ; Chitaley et al , ; Goswami et al , ; Kim et al , ; Goswami et al , , ; Goswami et al , ). Arachidonic acid cytochrome P450 epoxygenase pathway has been highlighted for its role in the relaxation of smooth muscle and vasodilation (Jin et al , ; Yousif and Benter, ).…”
