Establishing a mouse model of choroidal neovascularization to study the therapeutic effect of levotinib and its mechanism

Abstract: Objective To study the therapeutic effect and mechanism of levotinib on choroidal neovascularization (CNV) in mice. Methods 45 healthy C57BL/6 mice were selected and randomly divided into three groups: control group (group A), model group (group B) and levotinib group (group C). The model of CNV in mice was established. The fluorescence leakage of choroidal lesions in mice was observed by fundus fluorescein angiography. The morphological changes of retinal vessels in mi… Show more

“…Laser-induced CNV lesions were induced following a standard protocol ( Xin et al, 2020 ). In brief, animals were first anesthetized by intraperitoneal injection of avertin (2,2,2-tribromoethanol) (0.2 ml/10 g) (Nan Jing AIBI Bio-Technology Co., Ltd, Nanjing, China).…”

“…Hematoxylin and eosin (H&E) staining was performed according to a previously described procedure ( Wei et al, 2018 ; Xin et al, 2020 ). Mice were killed, and the eyes were enucleated 3°days (for safety test) or 7°days (for CNV volume analysis) after laser injury and fixed in 4% paraformaldehyde at 4 ° C for 24°h, conventionally dehydrated, and embedded in paraffin wax.…”

“…Laser burns were given carefully to avoid large vessels, but as close as possible to the optic nerve head (around 500 µm, see Section 3). Laser-induced disruption of Bruch's membrane was identified by the appearance of a bubble at the site of laser lesion [19][20][21]. In 16% of the cases, the laser spot was, however, localized more peripherally due to the anatomy of the vascular bed.…”

“…Before the test, mice were given an intraperitoneal injection of 5% chloral hydrate (10 mL per kg body weight) to produce hypnosis. 29,30 Then, 1% tropicamide eye drops were dropped into the eye for mydriasis, and levofloxacin transparent gel was used to protect the cornea.…”
