Establishing and maintaining cell polarity with mRNA localization in Drosophila

Abstract: How cell polarity is established and maintained is an important question in diverse biological contexts. Molecular mechanisms used to localize polarity proteins to distinct domains are likely context-dependent and provide a feedback loop in order to maintain polarity. One such mechanism is the localized translation of mRNAs encoding polarity proteins, which will be the focus of this review and may play a more important role in the establishment and maintenance of polarity than is currently known. Localized tra… Show more

“…Promoter-driven loading of Paip2 could be a way to mark specific transcripts involved in RNA-depending tissue-specific events, e.g. cell polarization [43], neuroplasticity [44], etc. Another possibility, not excluding the aforementioned one, is the involvement of Paip2 in some event(s) at the promoter.…”

“…Once the transcripts are on site, RNAbinding proteins interact with them to regulate their translation. The CPEB protein family is a group of translation factors that help anchor and control the on-site translation of localized transcripts [35][36][37][38][39][40].…”

“…Once the transcripts are on site, RNA-binding proteins interact with them to regulate their translation. The CPEB protein family is a group of translation factors that help anchor and control the on-site translation of localized transcripts [3540]. CPEBs recognize CPE elements in the 3’UTR of localized transcripts and can function to repress or activate their translation, depending on the context.…”

“…In neurons, the axons contain ribosomes and the translational machinery necessary for local translation of a specific subset of mRNAs [51]. While mRNA transport to specific localization of subcellular domains ultimately rely on a pre-existing cell polarity, on-site translation is efficient to generate and stabilize protein asymmetry [52]. With regard to polarity proteins, local translation of Pard3 in the growth cone has been observed in response to NGF and Netrin-1 signaling and to promote axonal elongation in neurons in culture [53].…”
