Establishment and application of population pharmacokinetics model of vancomycin in infants with meningitis

Abstract: Background: To establish a population pharmacokinetics (PPK) model of vancomycin (VCM) for dose individualization in Chinese infants with meningitis. Methods: We collected the data of 82 children with meningitis in hospital from July 2014 to June 2016. The initial vancomycin dosage regimen for children was 10 or 15 mg/kg for q12 h, q8 h or q6 h. Serum concentrations were determined by Viva-E Analyzer before and after the fifth administration. The PPK model was developed by nonlinear mixed-effect model software… Show more

“…Moreover, we studied the NEAB's selectivity against relevant interfering species. Figure S7 (B and C) shows that the tested biosensors' responses are minimally affected (<5%) by the presence of common endogenous molecules and frequently coadministered drugs [here, ibuprofen and azithromycin; also relevant to vancomycin monitoring (44)(45)(46)(47)].…”

“…Included in this review were eight population PK studies gathering data from a total of 2523 patients with an average of 9% (N = 228) receiving a loop diuretic [ 29 , 30 , 31 , 32 , 33 , 34 , 35 , 36 ]. The median and range of age, weight, and eGFR across the studies were 26.65 years (0.65–65), 65.55 kg (2.17–88), and 98.97 mL/min (39.24–120.6), respectively.…”

“…However, the guidelines mention that this was a weak recommendation, and the quality of evidence was very low. Vancomycin doses lower than the dose recommended by the IDSA have been observed in some studies of Chinese infants 31–33 . The dosage of 60–80 mg/kg/day may not be needed for Chinese infants with normal renal function.…”

“…Vancomycin doses lower than the dose recommended by the IDSA have been observed in some studies of Chinese infants. [31][32][33] The dosage of 60-80 mg/kg/day may not be needed for Chinese infants with normal renal function. The efficacy and safety of higher dosages should be further evaluated in Chinese infants with normal renal function.…”
