Establishment of three-dimensional cultures of human pancreatic duct epithelial cells

Abstract: Three-dimensional (3D) cultures of epithelial cells offer singular advantages for studies of morphogenesis or the role of cancer genes in oncogenesis. In this study, as part of establishing a 3D culture system of pancreatic duct epithelial cells, we compared human pancreatic duct epithelial cells (HPDE-E6E7) with pancreatic cancer cell lines. Our results show, that in contrast to cancer cells, HPDE-E6E7 organized into spheroids with what appeared to be apical and basal membranes and a luminal space. Immunostai… Show more

“…Many types of epithelial cells form organoid structures in 3D culture24,40,124126, recapitulating varying levels of tissue polarity and architecture. For MDCK cells, at least, this is analogous to a mesenchymal–epithelial transition (MET).…”

“…Pancreatic endocrine tissue has been cultured as organotypic slices from perinatal mouse embryos 27 and pancreatic cancer cell lines have been cultured on type I collagen-glycosaminoglycan scaffolds 28 and in Matrigel. 29 Little attention hitherto has been paid to the likely contribution of stromal cells, such a characteristic of pancreatic cancer, to tumor cell behavior.…”

“…Looking at these differences, though, it may not be simply a question of which study may be right or how both could fit into a common model. The difference could be the result of dose-dependent effects and biological responses between different cell lines or sublines thereof, which may be further complicated by the existence of a nuclear pool of CTMP in certain cell types (18).…”
