Ethanol potentiates the genotoxicity of the food-derived mammary carcinogen PhIP in human estrogen receptor-positive mammary cells: mechanistic support for lifestyle factors (cooked red meat and ethanol) associated with mammary cancer

Abstract: Consumption of cooked/processed meat and ethanol are lifestyle risk factors in the aetiology of breast cancer. Cooking meat generates heterocyclic amines such as 2-amino-1-methyl-6-phenylimidazo[4,5-b]pyridine (PhIP). Epidemiology, mechanistic and animal studies indicate that PhIP is a mammary carcinogen that could be causally linked to breast cancer incidence; PhIP is DNA damaging, mutagenic and oestrogenic. PhIP toxicity involves cytochrome P450 (CYP1 family)-mediated metabolic activation to DNA-damaging spe… Show more

“…In contrast to our hypothesis that anthropometric factors would influence tumor levels of AhR and aromatase, only a marginal association between WHR and AhR was found. Also, a marginal inverse association with alcohol abstention ( 49 ) but not smoking ( 7 ) was observed. Younger and nulliparous patients were substantially more likely to have tumors positive for intratumoral aromatase, both factors associated with ER – status ( 50 ).…”

“…The term “genotoxicity” refers to deleterious genetic changes involving mutations, aberrations in chromosomes caused by genotoxins. These genotoxins have the ability to modify the DNA sequence leading to several genetic abnormalities (Malik et al., 2018).…”

“…A recent study proved that only ROS concentrations coming from certain types of chinese cooking exhibited consistency with cell viability, and showed significant correlations with genetic damage and expression in human bronchial epithelial cells 38 . PhIP might be generating ROS by more than one mechanism, which would induce DNA damage and cell toxicity 39 . ROS was generated from MeIQx and the ROS might be involved in mutagenesis 40 .…”
