Ethanolic Neem (Azadirachta indica) Leaf Extract Prevents Growth of MCF-7 and HeLa Cells and Potentiates the Therapeutic Index of Cisplatin

Abstract: The present study was designed to gain insight into the antiproliferative activity of ethanolic neem leaves extract (ENLE) alone or in combination with cisplatin by cell viability assay on human breast (MCF-7) and cervical (HeLa) cancer cells. Nuclear morphological examination and cell cycle analysis were performed to determine the mode of cell death. Further, to identify its molecular targets, the expression of genes involved in apoptosis, cell cycle progression, and drug metabolism was analyzed by RT-PCR. Tr… Show more

“…The combination of neem-derived gedunin and cisplatin further decreases the proliferation of treated ovarian cancer cells by almost 50% compared to the cells treated with only cisplatin [28]. The combination of sub-lethal dose of ethanolic NLE and cisplatin also provides synergistic effects in decreasing the viability of breast and cervical cancer cells compared to individual compound alone [100]. …”

“…The susceptibility of tumor cells to MEX may be associated with the modification of the plasma membrane lipids during transformation of normal cells to malignant cells. The ethanolic neem leaf extract also specifically targets the breast as well as cervical cancer cells, without showing significant effects on the viability of primary lymphocytes [100]. …”

“…Manasathien et al (2012) assayed the cytotoxic activity of fruit peel extract of P. granatum on Brine shrimp and determined CC-50 of 1205.98 and 1294.88 lg/ml with ethanolic and water extracts respectively. Sharma et al (2014) reported EC-50 of ethanolic neem leaf extract for MCF-7 cells at 350 lg/ml after 72 h treatment whereas in HeLa cells it was found to be175 lg/ ml in 48 h.…”

“…Studies have shown that A. indica has got a chemo preventive capability by regression of the DEN/AAF (diethyl nitrosamine/2-acetylaminofluorene) induced carcinogenesis in liver [14,15]. These results indicate that dietary use of extracts from various parts of A. indica may play a promising role in future drug discovery and development programs as far as chemoprevention of cancer is concerned.…”
