Eukaryotic ribosome biogenesis at a glance

Abstract: Summary Ribosomes play a pivotal role in the molecular life of every cell. Moreover, synthesis of ribosomes is one of the most energetically demanding of all cellular processes. In eukaryotic cells, ribosome biogenesis requires the coordinated activity of all three RNA polymerases and the orchestrated work of many (>200) transiently associated ribosome assembly factors. The biogenesis of ribosomes is a tightly regulated activity and it is inextricably linked to other fundamental cellular … Show more

“…These assembly and processing events are tightly coupled and occur within preribosomal particles (see Glossary) that travel, as maturation progresses, from the nucleus across nuclear pore complexes (NPCs) to the cytoplasm, where they are ultimately converted into translation-competent ribosomal subunits [5,[9][10][11][12][13] (Figure 2, Key Figure). Given the gargantuan complexity of this process, it is unsurprising that the assembly of eukaryotic ribosomes strictly requires the assistance of a plethora (>200) of mostly essential ribosome biogenesis factors, which are also called trans-acting or assembly factors [5,14,15].…”

“…At the beginning of this millennium, the implementation of powerful in vivo affinity-purification approaches permitted the isolation and characterization of preribosomal particles [19,20]. Subsequently, the purification of additional preribosomal particles provided a detailed spatiotemporal picture of the preribosomal intermediates and their composition [5,9,14] (Figure 2). A few of these preribosomal particles were visualized at relatively low resolution by EM [21][22][23][24].…”

“…Ribosome biogenesis is a complex and tightly regulated process requiring the activity of all three RNA polymerases and the synthesis and coordinated assembly of multiple proteins and specific rRNAs (9). Thus, ribosome biogenesis requires a large bioenergetic investment (10) and is actively maintained in continuously proliferating, transformed cells and cell lines.…”

“…The rDNA locus exists as a tandem-repeat array comprising B150 rDNA units containing the 35S and 5S genes, which are transcribed by RNA polymerase I (PolI) and PolIII, respectively. Processing of the 35S pre-rRNA generates 5.8S, 18S and 25S rRNA that, together with the 5S rRNA, become the catalytic backbones of each ribosome 5,6 . Only in anaphase does yeast repress rDNA transcription 4 , which allows the sister rDNA loci to condensate and segregate.…”
