Evaluation of a Novel Reporter Virus Neutralization Test for Serological Diagnosis of Zika and Dengue Virus Infection

Abstract: Currently, the laboratory diagnosis of Zika virus (ZIKV) infection is primarily through the detection of ZIKV RNA or antibodies against ZIKV proteins. The detection of viral RNA is highly sensitive and specific, but periods of viremia and viruria are brief, limiting the utility of ZIKV RNA assays. Instead, most ZIKV infections are diagnosed serologically, using an IgM antibody capture enzyme-linked immunosorbent assay (MAC-ELISA) for screening, followed by a confirmatory plaque reduction neutralization test (P… Show more

“…PRNT 90 titers are subsequently compared between ZIKV and DENV to determine the specificity of the NAs (if present), although interpretive challenges exist with this method, as discussed below. Studies to improve on the classic PRNT technique are ongoing, including most recently the use of luciferase-labeled viruses to decrease the TAT while maintaining the accuracy of the classic PRNT (43). Importantly, however, while ZIKV testing guidance established by the CDC has been fluid, with multiple revisions over the past 2 years, the recommendation that positive results from anti-ZIKV IgM screens be confirmed by supplemental testing with the PRNT has remained constant (44).…”

“…This hypothesis was confirmed by ELISA assays, which did not recognize specific immunoglobulins in any of the 13 available sera against any of the tested viruses (ZIKV, DENV, YFV) ( Table 2). It is known that serum quality, especially the degree of hemolysis, directly influences serological tests [65,66]. Nevertheless, we decided to test all the samples, including the hemolyzed, because they were the only available material obtained from wild animals that rarely can be examined.…”

“…A total of forty de-identified convalescent sera from COVID-19 patients (confirmed with viral RT-PCR positive) were tested in this study. Ten non-COVID-19 sera, collected before COVID-19 emergence 12,13 , were also tested in the reporter virus and PRNT assays. For testing cross reactivity, a total of 138 de-identified specimens from patients with antigens or antibodies against different viruses, bacteria, and parasites were tested in the mNeonGreen SARS-COV-2 neutralization assay (Table 1).…”
