“…A drill-hole injury was created in animals as previously described [4, 9, 10]. Briefly, the front skin of the mid femur in rats was incised straight and longitudinally at 1 cm in length under anesthesia.…”

“…Generally in modern medicine, treatment of bone fractures involve the use of a cast or plaster to immobilise the fractured limb, followed by the administration of analgesics (pain killers) and anti-inflammatory drugs to patients. The broken bone is then allowed to regenerate naturally on its own [3], since there is no orally active pharmacological agent available for rapid fracture repair [4]. Even though fractures heal naturally, it would be of great interest to discover alternatives to accelerate a bone healing process, thereby reducing the healing period, reducing the cost of treatment and helping patients to quickly regain their good health.…”

“…A drill-hole injury was created by inserting a drill bit with a diameter of 0.8 mm in the femur metaphysis region following our previously described protocol with minor modification with respect to drill-hole spot [30]. On the fourteenth day, rats were killed and bones appropriately preserved for histology, histomorphometry and micro-computed tomography (μCT) following our previously described methods [31]. Twenty-four hours before killing, all animals received calcein (20 mg/kg) by intraperitonial route.…”

“…Trabecular bone volume (BV/TV; percentage) and trabecular number (Tb.N) of femur epiphysis and proximal tibial metaphysis were calculated by the mean intercept length method. Trabecular thickness (Tb.Th; mm) was calculated according to the method of Hildebrand and Ruegsegger (Ngueguim et al, 2012). 3D parameters were based on analysis of a Marching cubes-type model with a rendered surface.…”
