Evaluation of chemopreventive effects of Thymoquinone on cell surface glycoconjugates and cytokeratin expression during DMBA induced hamster buccal pouch carcinogenesis

Abstract: The present study aimed to investigate the membrane stabilizing effect of Thymoquinone (TQ) on cell surface glycoconjugates and cytokeratin expression against DMBA induced hamster buccal pouch carcinogenesis. 0.5% DMBA painting (three times per week) in hamster buccal pouches for 14 weeks resulted in the formation of well developed oral squamous cell carcinoma. We observed 100% tumor formation with marked abnormalities of glycoconjugates status in tumor bearing hamsters as compared to control animals. Oral adm… Show more

“…Chemoprevention is a novel and promising approach to control, inhibit or suppress the tumor promotion by using natural entities. A large number of phytochemicals ingested in human diet represent a chemopreventive agent which possess antimutagenic, anticarcinogenic, antioxidant and anti-cell proliferating properties (10).…”

“…These findings were in agreement with other studies (30,33) . Rajkamal et al (2010) (29) study indicates that TQ has potent chemopreventive efficacy in experimental oral carcinogenesis. Oral administration of TQ to DMBA painted hamsters significantly normalized and inhibited the abnormalities seen in cell surface glycoconjugates in the tumor tissues and circulation during carcinogenesis, and restored the expression of cytokeratin, which indicates their membrane stabilizing effect of TQ during neoplastic transformation.…”

“…Natural compounds are ubiquitously distributed throughout plants and marine organisms (23) . Chemoprevention is a promising approach to control, inhibit or suppress the tumor formation by using natural or synthetic entities, natural that possess antimutagenic, anticarcinogenic, antioxidant, anti-lipid peroxidative and anti-cell proliferating properties are considered as good chemopreventive agents in carcinogenesis including those of oral and experimental ones (6,(24)(25)(26)(27)(28)(29)(30) . A large number of phytochemicals ingested in human diet exhibit anticarcinogenic and antimutagenic effects (30).…”

“…The development of tumors in Hamster buccal pouch exhibits various similarities such as shape, size, pathological features, pre-tumor lesions, property to spread and destruct,, biochemical and molecular markers expression and alterations in genomes as expressed in human OSCC [31]. In Hamster buccal pouch carcinogenesis, a sequential progression is observed through various degrees of dysplasia that is from hyperplasia to invasive carcinoma [32]. In DMBA induced hamsters 100% formation of tumor occurrence, tumor mass, tumor size and tumor progress were noted and subsequently counterchecked by performing histopathological analysis in which conditions like hyperplasia, dysplasia and squamous cell carcinoma are revealed [33,34].…”
