Evaluation of different carbon sources for high frequency callus culture with reduced phenolic secretion in cotton (Gossypium hirsutum L.) cv. SVPR-2

Abstract: HighlightsWe have evaluated the role of carbon sources in controlling excessive phenolic secretion from callus culture of cotton.Maltose was found to control phenolic secretion when it is used as a sole carbon source.Efficient protocol was developed for high frequency callus induction and proliferation for SVPR-2 cotton cultivar using different plant growth regulators.Adjustment of carbon source concentrations was found to play an important role in cotton callus culture. The present work will be helpful for est… Show more

“…The results (Table 1) show that the combination of 0.1 mg dm −3 KT + 0.025 mg dm −3 IBA induced the highest differentiation rate of cotton. Our callus differentiation rate statistics for exogenously applied auxin and cytokinin were consistent with previous studies, i.e., the induction of EC and SEs, and the concentration of 2, 4-D in the medium was replaced by a combination of IBA and KT [24,37]. In this experiment, the role of auxin and cytokinin was not achieved alone, but by interacting with other pathways and components involved in the differentiation and development of SEs, such as their upstream TF regulators and hormone signaling pathways [38].…”

“…Rooting was declined vigorously when increasing (> 2.0 mg/L) concentrations of NAA. Similar results were also noticed in Sesamum indicum (Raja and Jayabalan, 2011), Solanum surattense (Rahman et al, 2011) and Gossypium hirsutum (Prem Kumar et al, 2015). …”

“…Other workers have also reported similar thick rosette types of shoots formation in higher concentrations of BA in case of Gossypium hirsutum (Prem Kumar et al, 2015; Commiphora wightii (Mohan et al, 2017). The shoot multiplication significantly varied according to the explant type.…”

“…In our experiment, maltose at a concentration of 90 g/L clearly reduced the L. aestivum plants' biomass. On the other hand, maltose at higher concentrations had a positive effect on F. indica and Gossypium hirsutum callus growth (Kumar et al, 2015;Khan et al, 2018).…”
