Evaluation of Dried Amniotic Membrane on Wound Healing at Split-Thickness Skin Graft Donor Sites: A Randomized, Placebo-Controlled, Double-blind Trial

Abstract: OBJECTIVE To evaluate the effect of amniotic membrane (AM) at split-thickness skin graft (STSG) donor sites. METHODS This double-blind randomized controlled trial was conducted on 35 eligible participants referred to the burn unit of Vasei Hospital of Sabzevar, Iran, during 2017 and 2018. Each STSG donor site was divided into two sides, and the respective halves were covered with either a dried AM or petrolatum gauze (control). Outcomes were evaluated o… Show more

“…The fact that the application of amniotic or amniochorionic membranes leads to a significant reduction in pain has been repeatedly noted in clinical studies using cryopreserved [ 31 , 51 , 52 ] or air-dried terminally sterilized tissue [ 53 ]. The analgesic effect of cryopreserved AM was mainly explained by the reduction of inflammation and thus secondary to pain relief [ 31 , 54 ].…”

“…To the best of our knowledge, no study has compared the analgesic effect between multiple types of AM. Of the studies where pain was assessed using a pain score, analgesic effect was verified in studies using fresh [ 16 , 17 ], cryopreserved [ 33 , 62 , 63 ], and air-dried terminally sterilized (gamma irradiation 25 k Gy) [ 34 ] AM allografts for various wound treatment.…”

“…During clinical studies, apart from accelerating healing, the most outstanding effect of AM application is rapid wound pain alleviation [ 17 , 33 , 34 ]. The exact mechanism of analgesic action of AM was not known for a long time and no directly acting component was characterized [ 35 ].…”
