Pokkah boeng, once a minor disease is becoming major disease of sugarcane a total of 14 bacterial isolates were obtained from sugarcane phylloplane using HiCrome Bacillus Agar and characterized culturally for colony colour, shape and number of colonies per plate. Grams' test differentiated the Gram-positive bacteria from Gram negative bacteria which was done using 3% KOH test. All these isolates were evaluated for their antagonism against Fusarium sacchari through dual culture technique. Out of 14 isolates, ScANKP-9 and ScANKP-13 isolates were found to be the most effective with maximum antagonistic potential by significantly restricting the growth of F. sacchari to 4.47 and 4.80 cm, in comparison to monocultured F. sacchari (9 cm) with 50.33 per cent and 46.67 per cent inhibition in radial growth. Molecular characterization of effective isolates using modified CTAB method following PCR amplification of DNA with FGPS-63 and FGPL-132-38 primers, BLAST and phylogenetic analysis confirmed the isolates, ScANKP-9 and ScANKP-13 as Bacillus amyloliquefaciens and Pseudomonas fluorescence, respectively.

O Brasil é o maior produtor de cana-de-açúcar e seus subprodutos do mundo, pois a cultura é muito favorecida pelas condições climáticas do país. Uma das preocupações e custos de produção para o setor sucroalcooleiro é a ocorrência de doenças durante todas as fases do cultivo da cana-de-açúcar. Dentre as principais doenças pode-se citar a podridão vermelha, causada pelo fungo Colletotrichum falcatum e a murcha e Pokkah Boeng causadas por diversas espécies de Fusarium spp. Essas doenças afetam o cultivo da cana em todo o mundo, reduzindo gravemente a produtividade da cana-de-açúcar na colheita. Provocam perdas de até 100% da produção e muitas variedades comerciais foram perdidas devido à severidade das doenças. Para podridão vermelha (C. falcatum) existem muitos estudos e variedades resistentes disponíveis, porém há poucos produtos e recomendações de manejo. Já para murcha e Pokkah Boeng (Fusarium spp.) existem diversos gargalos e informações que ainda não são conhecidas sobre a doença, incluindo as diferentes espécies do agente etiológico (Fusarium spp.) que podem causar a doença ou até outras doenças em que os sintomas são divergentes. A falta de produtos registrados para as doenças, sejam fungicidas químicos ou biológicos é outro desafio enfrentado, pois o manejo fica dependente apenas do desenvolvimento de cultivares resistentes, o que demanda grande investimento de tempo, além do financeiro. O presente artigo revisa o cenário da podridão vermelha, da murcha de Fusarium spp. e da Pokkah Boeng em cana-de-açúcar, o impacto das doenças no cultivo da cana-de-açúcar, características dos patógenos, resistência às doenças e manejo disponível.

The use of endophytic bacteria in agriculture provides an effective way of improving crop yield and significantly reducing chemical usage, such as fungicides. This research was conducted to explore endophytic bacteria with plant growth promotion (PGP) and antifungal activities against Fusarium moniliforme AIT01. In this study, we obtained 52 isolates of endophytic bacteria associated with the roots and stems of sugarcane from Nakhon Ratchasima province, Thailand. In vitro antagonistic activity test showed that 14 out of 52 isolates had antagonistic activity against the fungal pathogen F. moniliforme AIT01. These antagonistic endophytic bacteria were identified as belonging to six different species as follows: Nguyenibacter vanlangensis, Acidomonas methanolica, Asaia bogorensis, Tanticharoenia aidae, Burkholderia gladioli and Bacillus altitudinis based on phenotypic characteristics, along with phylogenetic analysis of their 16S rRNA gene sequences. Seven isolates effectively inhibited F. moniliforme AIT01 mycelial growth by up to 40%. The volatile compounds of six isolates reduced the growth of F. moniliforme AIT01 by over 23%. Moreover, riceberry rice seedlings previously treated with B. gladioli CP28 were found to strongly reduce infection with phytopathogen by 80% in comparison to the non-treated control. Furthermore, the isolates also showed relevant PGP features, including ammonia production, zinc and phosphate solubilisation, auxin and siderophore biosynthesis. These results demonstrated that the tested endophytic bacteria could be successfully utilised as a source of PGP and biocontrol agent to manage diseases caused by F. moniliforme.
