Evaluation ofPax5expression and comparison with BLA.36 and CD79αcy in feline non‐Hodgkin lymphoma

Abstract: Paired box gene 5 (Pax5) is a widely used B-cell marker for human and canine non-Hodgkin's lymphoma (nHL); however, in the literature there is only one case report using Pax5 in a cat B-cell lymphoma. The purposes of this study were to investigate the expression and detection of B-cell specific activator protein (BSAP) using a monoclonal anti-Pax5 antibody in feline nHL (FnHL) tissue samples to evaluate its diagnostic relevance as a B-cell marker. A total of 45 FnHL samples in 45 cats were evaluated. B-cell ly… Show more

“…This is in contrast with human MCC, which often have immunoexpression of PAX‐5 6,19,20,30,31 . Although, PAX‐5 is not widely used as a B‐cell marker in cats, it has been reported to result in similar staining as in human and canine samples 36 . KIT expression was positive in all cases examined, which is in line with previous findings in canine and human MCC 16,18,34,35 .…”

“…It uses labelled antibodies to stain for the expression of specific cell membrane receptors found on different lymphocytes. Commonly used antibodies include CD3 for T cells, and CD79 for B cells (Waly and others 2005, Moore and others 2012), and a recent study has evaluated Pax5, a protein that is important in B cell differentiation, as an additional useful marker for B cell lymphoma (Felisberto and others 2017). This technique allows the pathologist to confirm that an identified round cell population is lymphocytic and shares the same phenotype (monomorphic population).…”

“…26,37,38 Other B-cell antigens have been utilized in more recent studies with evidence of increased specificity, including PAX5. 12,40 Protein expression of multiple myeloma oncogene 1 (MUM1), also called interferon regulatory factor 4 (IRF4), has been utilized with other T-and B-cell antigens in dogs and cats in the diagnosis of plasma cell tumors. 27 There are rare reports of lymphoproliferative diseases in which B-and T-cell antigens are co-expressed in domestic animals, and the panel of antibodies used is often variable and limited.…”
