Evaluation of laser speckle contrast imaging as an intrinsic method to monitor blood brain barrier integrity

Abstract: The integrity of the blood brain barrier (BBB) can contribute to the development of many brain disorders. We evaluate laser speckle contrast imaging (LSCI) as an intrinsic modality for monitoring BBB disruptions through simultaneous fluorescence and LSCI with vertical cavity surface emitting lasers (VCSELs). We demonstrated that drug-induced BBB opening was associated with a relative change of the arterial and venous blood velocities. Cross-sectional flow velocity ratio (veins/arteries) decreased significantly… Show more

“…Another possible mechanism is that the increased vessel strain during larger SLEs results in an increase in plasma (or simply water) crossing the vascular epithelium. This is similar to the mismatch in arterial versus venous flow which we previously demonstrated using LSCI in a model of blood-brain barrier (BBB) leakage [23]. The results from LSCI-ECG synchronization presented here suggest an opportunity to measure more subtle changes in BBB permeability.…”

“…Among in vivo monitoring methods, magnetic resonance imaging (MRI) and computed tomography (CT) scanners are employed for monitoring the BBB permeability . However, these techniques suffer from a low resolution and sensitivity; in addition, they are very expensive to operate and not optimized for prolonged measurement periods . In contrast, various optical imaging techniques, such as contrast‐enhanced photoacoustic micro‐imaging , laser speckle contrast imaging and fluorescent imaging , have been developed for visualization of the BBB permeability in preclinical studies on rodents.…”

“…Quantification of motion is achieved by mathematically analyzing the spatial blurring [27,28]. A recent study [29] employed laser speckle contrast imaging to assess BBB permeability, constituting the first demonstration of LSCI in that aspect. The basis for this modality is that BBB disruption can alter cerebral blood flow [23].…”
