Evaluation of Mitotic Figures and Cellular and Nuclear Morphometry of Various Histopathological Grades of Oral Squamous Cell Carcinoma: Comparative study using crystal violet and Feulgen stains

Abstract: The Feulgen stain is more reliable than 1% crystal violet in terms of the selective staining of MFs. Moreover, the findings of this study indicate that computer-based morphometric analysis is an effective tool for differentiating between various grades of OSCC.

“… 28 An increase in mitotic perimeter directly indicates CIN, as it reflects irregular mitotic division, often resulting in chromosomal rearrangements and potential genetic abnormalities. 29 Here, the mean perimeter of metaphases was higher in LS mucCRC as compared to LS mucADE and increased along with increasing malignancy of the samples ( Fig. 2 ).…”

“…Since chromosomal instability manifests as atypical mitoses, 28 we reasoned it could be demonstrated by measuring the size of mitoses as previously described by Kesarkar et al. 29 Feulgen staining with light green background stain was used as previously described 12 to visualize nuclear material and mitoses in 4 carcinoma (LS_CAR2, LS_CAR3, LS_CAR4, LS_CAR5), 3 adenoma (LS_ADE9, LS_ADE11, LS_ADE12), and 5 colonic mucosa samples from patients with LS diagnosed either with adenoma (LS_N9, LS_N23) or carcinoma (LS_N5, LS_N6, LS_N7) ( Supplementary Table S1 ). A sigmoid colon carcinoma previously confirmed as MMR proficient (pMMR) and CIN positive by comparative genomic hybridization study (case no.…”

“…A proliferação excessiva de células devido ao aumento da mitoses atípicas é característica em lesões neoplásicas e pré-neoplásicas. Assim, avaliar quantitativamente o número de mitoses pode servir como um bom fator de prognóstico para tais lesões (Kesarkar et al 2018). A contagem de mitoses tem se revelado um importante fator na avaliação prognóstica para cães portadores de mastocitomas cutâneos (Romansik et al,2007).…”

“…The presence of mitotic activity and perineural invasion, as noted in our case, indicates a more aggressive behaviour and necessitates a comprehensive treatment approach. 10,11 Treatment strategies for MEC, particularly in minor salivary glands, revolve around surgical excision with clear margins, often supplemented by adjuvant radiotherapy for intermediate to high-grade tumors or those with adverse features. The therapeutic rationale parallels our management approach, advocating for a multidisciplinary strategy to optimize patient outcomes.…”
