IntroductionPistacia lentiscus L., which is also known as the lentisk or mastic tree, is widespread from Asia Minor to the Canary Islands and throughout the Mediterranean region (Ak and Parlakcı, 2009). It is an evergreen shrub with lovely red berries and dark green foliage and is often used as an ornamental plant (Mascarello et al., 2007). Lentisk can adapt to several climatic and pedological conditions (Zohary, 1952), such as severe drought (Correia and Catarino, 1994) and calcareous soil; it also exhibits regrowth after forest fires or deforestation (Ladd et al., 2005).P. lentiscus is also the source of a unique resin called mastic gum or mastic. It is obtained by wounding the trunk and thick branches of the male trees, since the females produce low levels of mastic (Acar, 1988). The cultivated male trees and mastic production have been exclusively maintained on the Greek island of Chios for a long time. Mastic production is also carried out in the adjacent Çeşme Peninsula of İzmir, Turkey, where ecological conditions are similar to Chios, Greece; however, mastic production in Çeşme is very restricted and does not match the quantity of the production on Chios Island (Baytop, 1968;Isfendiyaroglu, 2000). The agricultural activities and tourist facilities established in Çeşme have resulted in the continuous loss of mastic trees (Dogan et al., 2003). Therefore, the Turkish Foundation for Combating Soil Erosion for Reforestation and the Protection of Natural Habitats has been leading a project to protect the native mastic trees and establish new plantations on the Çeşme Peninsula to revive viable commercial production (www. tema.org.tr). In addition, a forest fire recently occurred on Chios Island and destroyed a total of 12,740 ha, which resulted in massive damage not only to the island's agricultural economy, but also to the world supply of Chios mastic (http://effis.jrc.ec.europa.eu). Mastic gum has a long tradition in folk medicine and has been used by healers for several treatments, such as for hypertension, coughs, sore throats, eczema, stomachaches, kidney stones, and jaundice (Ljubuncic et al., 2005b;Gardeli et al., 2008).The essential oil of mastic contains several secondary metabolites such as myrcene, limonene, terpinen-4-ol, alpha-pinene, beta-pinene, alpha-phellandrene, sabinene, para-cymene, and gamma-terpinene (Castola et al., 2000). Different nutrient media (MS [Murashige and Skoog 1962]; QL [Quoirin and Lepoivre 1977] and WPM [Lloyd and McCown 1980]); plant growth regulators BA (benzil adenin), GA 3 (gibberellic acid), IBA (indole-3-butyric-acid), NAA (naftalen acedic acid); and sucrose concentrations were studied to determine the in vitro culture effects on healthier and faster seedling development from mature lentisk (Pistacia lentiscus L.) seeds. After 28 days of culture, the percentage of germinated seeds was the highest (70%) in the full-strength MS medium. The cytokinin BA was superior to other tested treatments in terms of its ability to promote germination of lentisk seeds. When tested at different concentrations, sucrose gave the best results obtained at concentrations of 1-4%, whereas high concentrations (6 and 8%) mainly decreased germination rate and there was no a regular pattern for elongation of the aerial parts of plants. With this described protocol, on average 76.67% seeds germinated 4 weeks after culture. Developed seedlings were satisfactorily acclimatized in sterilized peat, soil and perlite containing compost, with high percentage survival viability was obtained 9 months after transfer to in vivo conditions (93.33%). The results obtained showed that the enriched full-strength MS medium supplemented with 1 mg L-1 BA and 3% sucrose induced homogeneous and healthy seedling development in a period of 4 to 8 weeks of culture.

The spring-summer photosynthetic behaviour of Pistacia lentiscus, a spontaneous evergreen bush of the Mediterranean macchia, was followed during two consecutive years. Outdoor measurements were carried out monthly in the period from May to September in the years 2004 and 2005. Mean values of net photosynthesis (P N ) of external tests show a typical daily trend: a rise until the maximum followed by a decline of assimilation with lower values maintained until the end of day. External data were validated by light-response curves, obtained under different thermal regimes in controlled conditions. External trials and measures in controlled conditions confirm an elevated photosynthetic activity below 30°C and a decrease over that limit. The results obtained evidence that the CO 2 assimilation of the P. lentiscus is influenced by stressful temperatures. The ecophysiological response to this limiting factor is an adaptation that concentrates on the photosynthetic activity in the hours of the day and in the periods of the year in which temperatures are more favourable.
