Evaluation of spike protein antigens for SARS-CoV-2 serology

Abstract: Background Spike protein domains are being used in various serology-based assays to detect prior exposure to SARS-CoV-2 virus. However, there has been limited comparison of antibody titers against various spike protein antigens among COVID-19 infected patients. Methods We compared four spike proteins (RBD, S1, S2 and a stabilized spike trimer (ST)) representing commonly used antigens for their reactivity to human IgG antibodies using indirect ELISA in serum from COVID-1… Show more

“…The spike protein elicits the strongest humoral response upon infection (24)(25)(26)(27) and should be used as a reference in populationbased seroprevalence studies (28). Spike protein ectodomains were obtained from the National Research Council of Canada for the following strains: Wuhan-Hu-1 (PRO1-429 (SmT1-1), B.1.617.2 (PRO7176-1 [SmT1(B.1.617.2]), and P.1 (PRO6875-2 [SmT1v3-B]), where they were produced according to protocols reported elsewhere (29)(30)(31)(32).…”

“…Many vaccines were developed for emergency use [ 2 ]. The vaccines could lead to the development of spike-specific IgG antibodies against SARS-CoV-2, so serology assays have been used to detect the spike protein domain antibodies induced by vaccination or prior viral exposure [ 3 ].…”

“…The objective of RT-PCR tests is to make a reagent react with the genetic code of the virus (RNA). The antigenic test focuses on the structural proteins of the virus, the spike protein (S), and the nucleocapsid protein (N) [45].…”
