Evaluation of the cytokine mannose-binding lectin as a mediator of periportal fibrosis progression in patients with schistosomiasis

Abstract: Introduction:We hypothesized higher mannose-binding lectin level and classic factors (i.e., age, sex, alcohol consumption, exposure, and specifi c treatment) are associated with the severity of periportal fi brosis in schistosomiasis. Methods: This crosssectional study involved 79 patients infected with Schistosoma mansoni with severe or mild/moderate periportal fi brosis. Serum concentrations of mannose-binding lectin were obtained by enzyme-linked immunosorbent assay (ELISA). Results: Higher serum level of m… Show more

“…In the northeastern of Brazil, Mannose-Binding Lectin which also acts as an opsonin released by mononuclear innate cells (monocytes, macrophages, dendritic cells), was reported to be a risk factor for advanced periportal fibrosis in chronic hepatic diseases as well as S. mansoni infection ( 73 ). This can be explained by the role of MBL as a central component of the innate immune response and its highly collagenous activity following host infection by invading pathogens ( 100 ).…”

“…The cytokines produced subsequently increase extracellular matrix that is important in the pathogenesis of liver fibrosis [25,26]. Similarly, higher serum levels of MBL have been found in patients with advanced periportal fibrosis due to schistosomiasis [27].…”

“…This protein plays an important role in activation of the complement system by binding the MBL/serine protease associated with MBL (MBL/MASP) and opsonization. MBL levels are higher in individuals with advanced periportal fibrosis 3,4 . Some polymorphisms identified in the promoter region of MBL2 -550 (H/L variant guanine to cytosine (G-C) nucleotide substitutions, rs11003125) and -221 (X/Y X/Y variant, C to G nucleotide substitution, rs7096206) and in the 5-untranslated region of exon 1 at position +4 (A/O variant, C to T nucleotide substitution (c.4T/C) or rs7095891) are responsible for functional alterations and influence the circulating concentration of MBL 5 .…”
