Evaluation of Toxicity and Gene Expression Changes Triggered by Oxide Nanoparticles

Abstract: Several studies have demonstrated that nanoparticles (NPs) have toxic effects on cultured cell lines, yet there are no clear data describing the overall molecular changes induced by NPs currently in use for human applications. In this study, the in vitro cytotoxicity of three oxide NPs of around 100 nm size, namely, mesoporous silica (MCM-41), iron oxide (Fe 2 O 3 -NPs), and zinc oxide (ZnO-NPs), was evaluated in the human embryonic kidney cell line HEK293. Cell viability assays demonstrated that 100 µg/mL MCM… Show more

“…It has been shown previously that there is an increase of these pro-inflammatory mediators from the kidney following exposure to various antigens [30,31]. In a similar theme exposure of human embryonic kidney cell line HEK 293 to 100 nm ZnO NMs resulted in up-regulation of IL6 and IL8 genes from the cells in vitro [32]. …”

“…12 Other, more recent reports demonstrate the use of these cells for investigating the potential toxicity of nanoparticles. [13][14][15][16] Andelman et al examined the effects of exposure to different types of Y 2 O 3 NPs on human foreskin fibroblast cells and demonstrated a concentration-dependent (25-500 µg/mL) cytotoxicity by live/dead cell assay. 10 To investigate the potential mechanisms of the Y 2 O 3 NPs toxicity, we exposed the HEK293 cells to different concentrations of NPs to first determine the acute cytotoxic dose (IC 50 ).…”

“…IC 20 values were calculated from dose–response curves and we performed DNA microarray analysis of HPAEpiC and HPPC cells exposed to IC 20 concentrations of the titanium NPs to identify the molecular toxicity signature of the selected NPs under study. The selection of IC 20 values was based on earlier studies …”
