Evaluation of Toxicity and Neural Uptake In Vitro and In Vivo of Superparamagnetic Iron Oxide Nanoparticles

Abstract: Superparamagnetic iron oxide nanoparticles (SPIO-NPs) have great potential to be used in different pharmaceutical applications, due to their unique and versatile physical and chemical properties. The aim of this study was to quantify in vitro cytotoxicity of dextran 70,000-coated SPIO-NPs labelled/unlabelled with rhodamine 123, in C6 glioma cells and primary hippocampal neural cells. In addition, we analyzed the in vitro and in vivo cellular uptake of labelled SPIO-NPs. The nanoparticles, with average size of … Show more

“…Conversely, Dextran-coated SPIONs (10–50 nm) showed to be safer, did not impair the hippocampal cell viability at low doses after 24 h of incubation [113], which confirm the positive effect of the polymeric coating [4,108], as also discussed in Section 4.3.…”

“…The majority of neurotoxicity studies on IONPs are performed in 2-dimensional in vitro cultures [83,85,109,112,113]. There is, however, an increasing interest on co-cultured and 3-dimensional cells cultures, which are more realistic models with respect to 2D systems [114,115], extended also to other types of inorganic NPs, such as silica NPs [116].…”

“…The in vitro biocompatibility experiments indicated a proper dosage during in vivo experiments. In our in vivo experiments, 5 nm USPIO nanoparticles were intravenously injected into rat at ∼1.8 mg·Fe/kg dosage, which could be translated into ∼26.4 μ g·Fe/mL blood volume [43]. Such dosage was well located in a safe range for a good biocompatibility and less nonspecific uptake by normal hepatic cells.…”

“…Certainly, IONPs toxicity depends on many factors such as size, shape, surface chemistry, and chemical compositions, which can be modified. For example, coating the IONPs with different substances such as PEG and dextran can efficiently reduce the toxic effect of IONPs (Khalid et al 2018;Pisanic Ii et al 2007).…”
