Evolutionarily Conserved E(y)2/Sus1 Protein Is Essential for the Barrier Activity of Su(Hw)-Dependent Insulators in Drosophila

Abstract: Chromatin insulators affect interactions between promoters and enhancers/silencers and function as barriers for spreading of repressive chromatin. The Su(Hw) protein is responsible for activity of the best-studied Drosophila insulators. Here we demonstrate that an evolutionarily conserved protein, E(y)2/Sus1, is recruited to the Su(Hw) insulators via binding to the zinc-finger domain of Su(Hw). Partial inactivation of E(y)2 in a weak mutation, e(y)2(u1), impairs only the barrier, but not the enhancer-blocking,… Show more

“…2). In agreement with a conserved function of the NPC as an insulator, the Su(Hw)-dependent insulator of the gypsy transposon often associates with the nuclear periphery and requires ENY2, the D. melanogaster Sus1 homologue, for its barrier activity (Kurshakova et al, 2007a), indicating a structural and active role of the NPC in isolating chromatin domains.…”

“…This DNA-binding protein establishes the chromatin insulator of the gypsy retrotransposon (Geyer et al, 1986;Parkhurst et al, 1988;Spana et al, 1988;Geyer and Corces, 1992;Dorsett, 1993). The function of the gypsy insulator requires Su(Hw) recruitment of Centrosomal Protein 190 kD (CP190) (Pai et al, 2004), Modifier of mdg4 67.2 kD isoform (Mod67.2) (Georgiev and Kozycina, 1996) and Enhancer of yellow 2 [E(y)2; also known as ENY2] (Kurshakova et al, 2007a). Mod67.2 and CP190 are required for enhancer blocking by the gypsy insulator (Georgiev and Gerasimova, 1989;Pai et al, 2004).…”

“…E(y)2 is a subunit of the SAGA histone acetylation complex and the TREX RNA export complex (Kurshakova et al, 2007b). E(y)2 is required for barrier function of the gypsy insulator (Kurshakova et al, 2007a), possibly through its chromatin-opening activity (Kurshakova et al, 2007b). The gypsy insulator displays a versatile capacity to define transcriptional domains, protecting transgenes from enhancer and silencer action when inserted randomly throughout the genome (Roseman et al, 1993;Roseman et al, 1995a;Roseman et al, 1995b;Markstein et al, 2008).…”

“…The 1A2 insulator located on the 3Ј side of the yellow gene contains two binding sites for the Su(Hw) protein. Additional proteins, Mod(mdg4)-67.2, CP190 and E(y)2 (Gerasimova et al, 1995;Pai et al, 2004;Kurshakova et al, 2007), interact with Su(Hw) and are required for the activity of Su(Hw)-dependent insulators. None of the known DNA-binding insulator proteins binds to the Wari insulator located on the 3Ј side of the white gene (Chetverina et al, 2008).…”
