Ex-vivo characterization of circulating colon cancer cells distinguished in stem and differentiated subset provides useful biomarker for personalized metastatic risk assessment

Abstract: BackgroundCirculating tumor cells (CTCs) represent one of the most interesting target in improving diagnosis, prognosis and treatment. Herein we evaluate the possibility of using an emo-cytometric approach on the evaluation of the heterogeneous population of CTCs to improve personalized metastatic risk assessment. We benchmarked ex vivo behavior of distinct subsets of circulating colon tumor cells with correspondent clinical behavior of patients from which we isolated CTCs.MethodsIsolation and CTC expansion we… Show more

“…The culture mediums were conditioned during a short-time cultivation of fourteen days performed as previously described [13]. This time is sufficient to enrich the culture medium of soluble fraction of proteins produced by cells [14,15]. Moreover, different metabolic condition and generation of free radicals can influence its electrolytic state.…”

“…Differential expression of CXCR4 is critical for the maturation and selection of B cells [26,27] and sustained by hypoxic gradient. The SDF1/CXCR4 is involved in the inflammation surrounding metastatic progression of many carcinomas [28] comprising the NCDTs like in schwannoma [29].…”

“…Improvements have been made concerning the use of molecular profiling to guide the therapeutic intervention across a broad range of pathological conditions in the field of oncology, cardiology and ophthalmology, and chronic kidney disease (CKD) . Patient stratification was performed on the basis of proteome analysis alone, or in combination with multi ‐omics profiling data from tissue or body fluids.…”

“…However, the therapeutic benefit of the molecular‐guided treatment was not obvious . Malara et al targeted the assessment of the risk of metastasis of patients with colon cancer using circulating tumor cells (CTCs), supplemented with proteomics profiling . Based on the growth pattern in vitro, short‐time expanded CTCs could be clearly categorized into two subsets, i.e.…”
