Excess of early onset multiple myeloma in endometrial cancer probands and their relatives suggests common susceptibility

Abstract: Objective-Determine whether there is an association between uterine cancer and multiple myeloma.Methods-Data on second malignancies were obtained for 368 uterine corpus cancer patients treated between 1992 and 2005. Detailed family histories were devised for 192 probands. Diagnoses of multiple myelomas, lymphomas and leukemias in family members were medical record verified. The frequency of multiple myeloma among uterine cancer patients was compared to the female ageadjusted incidence rate of multiple myeloma … Show more

“…Hematologic and solid cancer risks also appear to be higher in relatives of multiple myeloma patients,8 especially chronic lymphocytic leukemia and non-Hodgkin lymphoma,4 as well as prostate cancer and endometrial cancer 8,14,16,17. Brown et al18 also have reported an increased risk of multiple myeloma associated with a family history of any hematologic cancer (odds ratio [OR] = 1.7) but did not find a significant increase for solid cancers in both white and black US families.…”

“…Adenocarcinoma of the colon is mostly described either before or after a diagnosis of MM [9]. Defective DNA mismatch repair and PTEN mutation have also been implicated in the progression of multiple myeloma, with possible inherited genetic susceptibility being suggested [10].…”
