“…The urine flow rate, which is markedly affected by saline loading [3,7-9.13,16], has a well-estab lished effect on urea clearance over the range of flow rates under consideration [10,12]. Thus, the failure of the BUN to rise after glycerol administration when a high urine flow rate is maintained by saline drinking may not indicate protection from acute renal failure, but rather the effect of the saline diuresis on the urea clearance and BUN con centration.…”

“…The conclusions were mainly based on blood urea nitro gen (BUN) determinations. The use of this indirect meas ure of GFR appears suboptimal on theoretical grounds considering that tissue damage accompanies the glycerol model of acute renal failure [I I], and that the saline diuresis associated with saline loading would be expected to alter urea clearance independent of the GFR [10,12]. More over, the reported protective effect of saline does not alter the renal morphologic changes that characterize glycerolinduced renal failure [8], We therefore reexamined the protective effects of saline loading on the glycerol model of acute renal failure using standard renal inulin clearance techniques to measure GFR.…”

“…Body weight averaged 218 ± 6 g. By previously described methods, polyethylene catheters were inserted in a carotid artery (for blood sampling and the measure ment of blood pressure using a transducer and a poly graph), a jugular vein (for fluid administration), and both ureters [15]. The left kidney was prepared for micropuncture as previously described [16], Following surgery, the rats were given an intra venous priming injection of inulin dissolved in isotonic NaCl. Plasma inulin concentration was maintained by continuous intravenous infusion of inulin in iso tonic NaCl given at 0.02 mi/min.…”
