Exosomal circRNAs: Emerging Players in Tumor Metastasis

Abstract: Metastasis is an important feature of malignant tumors, and is the primary cause of poor prognosis and treatment failure, in addition to representing a potentially fatal challenge for cancer patients. Exosomes are small extracellular vesicles 30–150 nm in diameter that transmit cargo, such as DNA, RNA, and proteins, as a means of intercellular communication. Exosomes play crucial roles in a range of human diseases, especially malignant tumors. A growing number of studies have verified that circRNAs can be enve… Show more

“…Tumor immunotherapy has been widely investigated and employed in clinical fields, as it holds great promise as a neoadjuvant therapy besides surgery and radiotherapy (61)(62)(63). At present, many inhibitors have been identified to play emerging roles in tumor suppression by regulating TME or assisting immuno-surveillance by immune cells (64).…”

“…Malignant tumor cells disseminate from the primary tumor site through the lymphatics, blood vessels, or body cavities to other body parts during metastasis [ 1 , 2 ]. This is a complex and multi-step process (Fig.…”

“…During tumor metastasis, cancer cells from the primary tumors spread through the circulatory system or body cavities to colonize distant organs. These tumor cells then further colonize novel sites to form metastatic sites in distant organs [ 1 , 2 ]. Various factors affect the occurrence and development of metastatic tumors, including genetic factors and in vivo microenvironment.…”

“…Exosomes of endosomal origin are nanoscale membrane vesicles (with a size of 30 ~ 100 nm) that coexist with microvesicles and apoptotic bodies in the extracellular microenvironment [ 83 ]. Exosomes can transfer various proteins, DNAs, and RNAs to mediate cell communication.…”
