Bacterial endosymbionts play essential roles in the biology of their arthropod hosts by interacting with internal factors in the host. The whitefly Bemisia tabaci is a worldwide agricultural pest and a supervector for more than 100 plant viruses. Like many other arthropods, Be. tabaci harbours a primary endosymbiont, Porteira aleyrodidarum, and an array of secondary endosymbionts that coexist with Portiera inside bacteriocyte cells. Unlike all of the other secondary symbionts that infect Be. tabaci, Rickettsia has been shown to be an exception by infecting insect organs and not colocalizing with Portiera, and has been shown to significantly impact the insect biology and its interactions with the environment. Little is known about the molecular interactions that underlie insect–symbiont interactions in general, and particularly Be. tabaci–Rickettsia interactions. Here we performed transcriptomic analysis and identified vitellogenin as an important protein that influences the levels of Rickettsia in Be. tabaci. Vitellogenin expression levels were lower in whole insects, but higher in midguts of Rickettsia‐infected insects. Immunocapture‐PCR assay showed interaction between vitellogenin and Rickettsia, whereas silencing of vitellogenin resulted in nearly complete disappearance of Rickettsia from midguts. Altogether, these results suggest that vitellogenin plays an important role in influencing the levels of Rickettsia in Be. tabaci.

Accurate detection and quantification of Candidatus Liberibacter solanacearum (CLs), the putative causal agent of zebra chip disease of potato (Solanum tuberosum L.), in the potato psyllid, Bactericera cockerelli (Sulc), has become necessary to better understand the biology of the disease cycle. Studies on the transmission efficiency of potato psyllids have shown inconsistencies with field surveys. There have also been reports of laboratory colonies inexplicably losing and regaining CLs infection as detected by polymerase chain reaction (PCR). Until now, DNA primers were used to detect CLs in potato psyllid tissue using conventional polymerase chain reaction (PCR) and gel electrophoresis or by real-time quantitative PCR. In this study, CLs was detected using bacterial tag-encoded FLX amplicon pyrosequencing (bTEFAP) at levels identifiable by PCR, and low levels, including samples with only one cell of CLs. Potato psyllids with <300 pyrosequencing reads did not show positive using conventional PCR. These results indicate that the currently accepted PCR diagnostic technique produces false negatives due to detection limits higher than what is generally present in field collected psyllids, and also provides an explanation as to why laboratory colonies seem to lose and regain CLs infection.

BackgroundRecent studies show that galling Hymenoptera and Diptera are able to synthesize the plant hormone indole-3-acetic acid (auxin) from tryptophan and that plant response to insect-produced auxin is implicated in gall formation. We examined the leaf transcriptome of galled and ungalled leaves of individuals of the Hawaiian endemic plant Metrosideros polymorpha (Myrtaceae) subject to infestation by psyllid (Hemiptera) gall-makers in the genus Trioza (Triozidae).ResultsTranscript libraries were sequenced using Illumina technology and the reads assembled de novo into contigs. Functional identification of contigs followed a two-step procedure, first identifying contigs by comparison to the completely sequenced genome of the related Eucalyptus, followed by identifying the equivalent Arabidopsis gene using a pre-computed mapping between Eucalyptus and Arabidopsis genes. This allowed us to use the rich functional annotation of the Arabidopsis genome to assess the transcriptional landscape of galling in Metrosideros. Comparing galled and ungalled leaves, we find a highly significant enrichment of expressed genes with a gene ontology (GO) annotation to auxin response in the former. One gene consistently expressed in all galled trees examined but not detected in any libraries from ungalled leaves was the Metrosideros version of SMALL AUXIN UPREGULATED (SAUR) 67 which appears to be a marker for leaf-galling in Metrosideros.ConclusionsWe conclude that an auxin response is involved in galling by Metrosideros psyllids. The possibility should therefore be considered that psyllids (like other insects examined) are able to synthesize auxin.
