Expression of d-psicose-3-epimerase from Clostridium bolteae and Dorea sp. and whole-cell production of d-psicose in Bacillus subtilis

Abstract: Purpose: D-psicose-3-epimerase (DPEase) catalyses the isomerisation of D-fructose to D-psicose, a rare sugar in nature with unique nutritional and biological functions. An effective industrial-scale method is needed for D-psicose production. Herein, the expression of a neutral and a slightly acidic pH DPEase in Bacillus subtilis was evaluated. Methods: Two DPEase genes from Clostridium bolteae and Dorea sp. were separately expressed in B. subtilis via plasmid pSTOP1622, and an extra P43 promoter was employed t… Show more

“…E. coli and B. subtilis WB800 transformants were incubated in LB medium containing 10 g/L peptone, 5 g/L yeast extract and 10 g/L NaCl, supplemented with 100 μg/mL ampicillin and 15 g/L agar as appropriate. Engineered B. subtilis cells were fermented in basal medium containing 10 g/L glucose, 15 g/L yeast extract, 1 g/L Na 2 HPO 4 , 1 g/L MgSO 4 ⋅ 7H 2 O and 8 g/L NaCl [46] . Recombinant E. coli and B. subtilis cells were cultivated in a 1 L flask containing 100 mL of fermentation medium at 37 °C with shaking at 200 rpm in shaking table (ZWYR‐240, Zhi Cheng Co., Ltd., Shanghai, China) [46] .…”

“…Usually, PHAs are made from bacteria that primarily consist of sugar or lipids as the intracellular product. About 250 different microorganisms are employed in the production of PHA (Mosca et al, 2020;Wei et al, 2020). In this process, bioplastics occur during the destruction of bacteria, specifically when separated from the microcells.…”
