Abstract. In the pig, the periimplantation period is critical for successful establishment of pregnancy. We studied the influence of embryos on oxytocin (OT) and progesterone (P4) regulated endometrial and myometrial secretion of 1) luteotrophic prostaglandin E2 (PGE2) and 2) luteolytic prostaglandin F2α and its metabolite (PGFM) on days 12-14 of pregnancy in pigs. We used unilaterally pregnant pigs created by a surgical procedure in which one uterine horn remained intact and the second horn was cut transversely so that part of the horn was detached from the uterine body. The animals were divided into two groups, inseminated gilts (days 12-14 of pregnancy, n=6) and uninseminated cyclic gilts, which were used as controls (days 12-14 of estrous cycle, n=5). Embryos developed only in the patent part of the uterus and not in the occluded horn. The abundance of OTR mRNA was increased in the endometrium and decreased in the myometrium of the gravid uterine horn in the pregnant pigs compared with the non-gravid uterine horn or either uterine horn in the cyclic pigs, indicative of a local effect of the conceptus. The presence of embryos in the uterine horn during the periimplantation period determines endometrial metabolism of PGF2α and the local response of the endometrium to OT and P4. OT stimulates PGF2α secretion and PGFM accumulation in endometrial cultures only from the non-gravid uterine horn and controls PGE2 secretion from the endometrium and myometrium in the pregnant gilts. The results indicate a more systemic affect of pregnancy on the uterine response to OT and a possibly the local effect of the conceptus in promoting progesterone's inhibition of OT-stimulated PGE2 secretion and uterine metabolism of PGF2α. Oxytocin, acting through oxytocin receptors (OTR) in the endometrium and myometrium, is involved in the control of prostaglandin F2α (PGF2α) and prostaglandin E2 (PGE2) secretion in pigs [3][4][5][6][7][8]. Prostaglandin F2α is responsible for luteal regression in non-pregnant pigs [1,9], while prostaglandin E2 acts as a luteotrophic and antiluteolytic factor that promotes maintenance of the corpus luteum during early pregnancy in pigs [10,11].Alteration of PG secretion by OT is implicated in maternal recognition of pregnancy in pigs [12][13][14]. Moreover, OT stimulates myometrial secretion of PGF2α and PGE2 as well as the contractile activity of the porcine myometrium during early pregnancy [8,15,16]. The present study was undertaken to determine the effect of interleukin-1β (IL-1β), interleukin-6 (IL-6) and tumour necrosis factor-α (TNF-α) on prostaglandin (PG)F 2α and PGE 2 secretion as well as cyclooxygenase-2 (COX-2) protein expression in myometrium collected on days 25, 30 and 40 of pregnancy in pigs. Myometrial slices were incubated for 16 h with IL-1β, IL-6 and TNF-α (1 or 10 ng/ml of medium) or two combinations of the three cytokines (1 or 10 ng/ml of each cytokine per combination). We demonstrated the stimulatory effect of IL-1β and IL-6 on PGF 2α and PGE 2 secretion from myometrium collected on all examined days of pregnancy, excepting of influence of IL-6 on release of PGF 2α by tissue from day 30. In turn, TNF-α was able to stimulate only PGE 2 secretion by myometrium of 40-day-pregnant gilts. The three cytokines applied in combination augmented release of PGE 2 from myometrium collected on days 30 and 40 of pregnancy. Stimulation of PGE 2 secretion by cytokines used individually was more frequent than that of PGF 2α . Moreover, an enhancement in PGF 2α and/or PGE 2 release was accompanied by an increase of COX-2 protein expression. Our study shows the ability of cytokines to stimulate PGF 2α and PGE 2 release by porcine myometrium from the first third of pregnancy. Obtained data suggest that locally PGs produced in myometrium influencing the uterine contraction activity may be important for the maintenance of myometrial quiescence during pregnancy and confirm also that the complex cytokine network is an important regulatory mechanism of PGs production during pregnancy.

Plasma was obtained for measurement of estradiol concentrations from D-2 to D0 and from D0 to D7 for progesterone measurements. On D7, the animals that ovulated in response to GnRH were slaughtered and uterus flushing and endometrium were collected. Differences between group means were determined by student′s t test. Animals from LF/CL group presented larger preovulatory follicles, corpus luteum, estradiol and progesterone levels when compared to animals from SF/CL group (P<0.1). The quantification of the transcripts was performed by qPCR.PTGES, AKR1C3, AKR1C4, CBR1, PTGIS and ALOX12 were more abundant and PLA2G10 was less expressed in the endometrium from animals from LF/CL when compared to SF/CL (P<0.10). Metabolites concentration was measured by mass spectrometry in endometrial tissue (pg/g) and uterine flushings (ng/mL). The metabolite 9-HETE was more abundant on uterine flushings from animals with SF/CL (P<0.05). Different concentrations of estradiol (proestrous) and progesterone (beginning of diestrous) modulated the expression of genes related to eicosanoids synthesis, but it did not affect eicosanoids levels in the endometrium and uterine flushing in beef cattle on day 7 of the estrous cycle.
