Expression of intercellular adhesion molecule-1 (CD54) on hematopoietic progenitors

Abstract: The distribution of intercellular adhesion molecule-1 (ICAM-1), a ligand for lymphocyte function antigen-1, on hematopoietic tissue was determined using the anti-ICAM-1 monoclonal antibody CL203.4 with flow cytometry and short-term semi-solid hematopoietic progenitor cultures. After timed incubation in media with fetal bovine serum, 29% of erythroid burst-forming units (BFU-E), 24% of erythroid colony-forming units (CFU-E), and 52% of granulocyte-macrophage colony-forming units (CFU-GM) bone marrow progenitors… Show more

“…Blast cells can constitutively express 1 and 2 integrins (Liesveld et al, 1993;Bendall et al, 1993) and Ig-like molecules (Maio et al, 1990;Arkin et al, 1991;Reuss-Berst et al, 1995). The expression of these proteins on leukaemic cells may be up-regulated by ATRA, and this might confer on blast cells a new adhesion potential.…”

“…EC activation by IL-1 or TNF-increases the expression of ICAM-1 and induces new EC adhesion molecules, like the endothelial leucocyte adhesion molecule-1 (E-selectin) and VCAM-1 (Mantovani & Dejana, 1989). ICAM-1 and VCAM-1 are also expressed by haemopoietic progenitors and leukaemic cells and may play a role in cell-cell interactions (Maio et al, 1990;Arkin et al, 1991;Reuss-Borst et al, 1995).…”

“…Unregulated proliferation and abnormal circulation in CML could therefore be caused by abnormal adhesive interaction between bone marrow stroma and malignant CML progenitors. Because this abnormal adhesive interaction may be caused by some alterations in adhesion molecules on CML progenitors, we investigated the expression of the adhesion molecules including CD44, VLA-5, VLA-4, LFA-1, ICAM-1, L-selectin and c-kit, previously described to be expressed on primitive haemopoietic progenitors (Kansas et al, 1990;Arkin et al, 1991;Gunji et al, 1992), of bone marrow CD34 cells from CML patients. received no treatment before study (two males and three females, 38-76 years old).…”

“…Consequently, there is a continuous remodelling of the cell surface of the differentiating cells and some membrane-expressed components are down-regulated whereas others are progressively acquired. It is well established, for instance, that adhesion receptors present on early committed progenitors (b1 and b2 integrins, CD54, CD44) are lost or severely decreased as the cells mature to erythrocytes, providing a potential mechanism for the release of mature erythroblasts from bone marrow into the peripheral blood (Kansas et al, 1990;Vuillet-Gaugler et al, 1990;Arkin et al, 1991;Papayannopoulou & Brice, 1992). Other receptors or cell surface markers, some being lineage-speci®c, become transiently or permanently expressed.…”
