Expression of single-chain antibody against RgpA protease of Porphyromonas gingivalis in Lactobacillus

Abstract: Aims:  The monoclonal antibody 61BG1·3, recognizing the RgpA protease, has been reported to confer protection against recolonization by the periodontal pathogen Porphyromonas gingivalis in humans. The aim of this study was to express a functional scFv derived from the monoclonal antibody 61BG1·3 on the surface of Lactobacillus paracasei for potential use in the prevention or treatment of periodontal diseases. Methods and Results:  The scFv was fused to an E‐tag and cloned in the Escherischia coli/Lactobacillus… Show more

“…Correct insertions of the VHH genes into the plasmids were verified by sequencing of the complete expression cassettes. The expression plasmids were transfected into L. paracasei BL23 by electroporation as previously described (23,32), generating strains of Lactobacillus expressing the anti-TcdB VHH antibodies, either secreted or cell wall displayed (see Table S2). …”

“…Lactobacilli are not existing usually in sulcus of gingiva but they can be formulated to show its antibacterial property on P gingivalis. Such a kind of method, organized by means of the innate immunomodulatory properties of probiotics, influence deal remarkable predictions in the coming years [13] .…”

“…In each cassette, the antibody fragment was also fused to an E tag for detection with an anti-E-tag antibody in Western blotting and enzyme-linked immunosorbent assay (ELISA). The pAF plasmid series (pAF100 to pAF1100) containing the 11 expression cassettes was introduced into L. paracasei (previously known as L. casei or L. zeae ATCC 393 pLZ15 Ϫ ) (7,15) by electroporation as previously described (7,10), generating L. paracasei pAF100 to pAF1100. The most important bacterial strains and plasmids used in this study are listed in Table 1.…”
