Expression of the Cydia pomonella granulovirus matrix metalloprotease enhances Autographa californica multiple nucleopolyhedrovirus virulence and can partially substitute for viral cathepsin

Abstract: The Cydia pomonella granulovirus open reading frame 46 (CpGV-ORF46) contains predicted domains found in matrix metalloproteases (MMPs), a family of zinc-dependent endopeptidases that degrade extracellular matrix proteins. We showed that CpGV-MMP was active in vitro. Autographa californica multiple nucleopolyhedrovirus (AcMNPV) expressing CpGV-ORF46 replicated similarly to a control virus lacking CpGV-ORF46 in cultured cells. The effects of AcMNPV expressing CpGV-MMP on virus infection in cultured cells and Tri… Show more

“…The proteins derived from VG008 OBs increased 27% the larvae mortality caused by SfCOL NPV, demonstrating that they are responsible for this enhancing activity. Several studies using recombinants proteins have also demonstrated the enhancer activity of these proteins in different granulovirus coinfection [13,25,35,37].…”

“…Whereas the loss of the cathepsin and chitinase genes is a common event among the betabaculoviruses [ 10 ], the matrix metalloproteinase gene is present in all betabaculoviruses sequenced to date [ 20 ]. The expression of a functional CpGV-encoded metalloproteinase into the Autographa californica multiple nucleopolyhedrovirus (AcMNPV) genome enhanced the virus virulence, promoted larval melanization, and could partially substitute for the viral cathepsin [ 21 ].…”

“…ODV initiates infection in the midgut cells of the host insects and is responsible for primary infection, while BV infects other tissues of the insect and is responsible for systemic or secondary infections [ 7 ]. Autographa californica multiple nucleopolyhedrovirus (AcMNPV), a model virus of the baculoviridae, has been extensively studied [ 8 , 9 , 10 , 11 , 12 , 13 ]. Further development of these applications requires further insight into the cell entry pathway.…”
